INDUCTION OF PROLIFEROUS PANCREATIC ISLET PRECURSOR CELL-LIKE CELLS BY TRANSIENT EXPRESSION OF MYCL AND INDUCTION OF DIFFERENTIATION INTO INSULIN-POSITIVE CELLS
20230173020 · 2023-06-08 · ·

Establishment of pancreatic islet-like insulin producing cells by inducing differentiation of ES/iPS cells has been reported. However, no technique has been developed so far for producing functional pancreatic islet insulin-positive cells in a large amount. In addition, there are concerns regarding rejection responses, accidental immune responses, etc. The present invention provides pancreatic islet-like cells having Mycl gene introduced thereinto and a method that comprises inducing proliferation of pancreatic islet-like cells by transient expression of Mycl gene and then inducing degradation thereof into insulin producing cells.

Inhibitors of miRNA-128 capable of promoting cardiomyocyte mitotic cell proliferation and methods effective for regeneration of heart tissue.

TRANSGENIC MOUSE FOR THE ASSESSMENT OF THYROID HORMONE (TH) ACTION

A transgenic animal model that is suitable for the cell or tissue specific assessing of thyroid hormone (TH) action in vivo is described. The recombinant DNA construct and methods suitable to generate such an animal are also provided. The assessment of TH action is based on a reporter that is dependent on an endogenously expressed thyroid hormone receptor (TR) and coregulators of said receptor.

ADAM12 inhibitors and their use against inflammation-induced fibrosis
09777276 · 2017-10-03 · ·

The present invention relates to the field of fibrosis and inflammation and more particularly to the use of ADAM12 (A Disintegrin and Metalloproteinase 12) inhibitors to prevent or treat inflammation-induced fibrosis. The present invention also relates to the use of ADAM12 as a marker for inflammation-induced fibrosis and to the ablation of ADAM12 expressing cells as therapeutic approach to interfere with the development of pro-fibrotic cells.

METHOD OF REDUCING NEURONAL MICROTUBULE BINDING PROTEIN TAU (TAU) LEVELS
20220031778 · 2022-02-03 ·

There is provided a method of reducing neuronal microtubule binding protein Tau (Tau) levels, promoting neuronal Tau degradation and/or promoting neuronal survival, in a subject in need thereof comprising contacting the subjects neurons with an effective amount of an agent that increases a long phosphotyrosine-binding (PTB) Numb isoform expression and/or activity, whereby neural Tau levels is reduced in the presence of the agent, the neuronal Tau degradation is promoted and/or the neuronal survival is promoted as compared to in the absence thereof. Also provided are methods of stratification based on PTB Numb isoform expression and/or activity of the subjects and compositions and kits for applying the methods.

TETRACYCLINE-INDUCIBLE EXPRESSION SYSTEMS
20220307056 · 2022-09-29 ·

The invention provides inducible promoter systems and their components incorporating components of a tetracycline operon. By coordinating expression of different transcriptional units in these systems as a result of selection of promoters and/or linking the units into the same DNA molecule, these systems can achieve higher levels of expression of coding segments of interest, increased differential levels of expression between on- and off-states, and/or greater responsiveness to inducing agents than conventional systems.

Recombinant AAV-crumbs homologue composition and methods for treating LCA-8 and progressive RP

The present invention relates to a Crumbs homologue (CRB) therapeutic for use as a medicament or in a method of treatment or prophylaxis, for example in the treatment or prophylaxis of a retinal disorder due to mutations in the Crumbs homologue-1 (CRB1) gene, such as Leber's congenital amaurosis 8 (LCA8) or retinitis pigmentosa 12 (RP12). In particular, the present invention relates to a recombinant viral vector comprising CRB2 or modified non-toxic forms of either CRB1 or CRB3 that resemble CRB2.

TRANSGENIC MOUSE MODEL FOR CONDITIONAL FKBP51 EXPRESSION AND RELATED METHODS

The subject invention pertains to transgenic non-human animals comprising a transgenic nucleotide sequence, integrated into the genome of the animals, comprising a nucleotide sequence encoding human FKBP51 operably linked to a tetracycline response element. In some embodiments, the transgenic animal comprises an additional transgenic nucleotide sequence, integrated into the genome of the animal, comprising a nucleotide sequence encoding a tetracycline transactivator (tTA) operably linked to a promoter; wherein the tTA is expressed upon activation of the promoter and binds the tetracycline response element, thereby causing expression of FKBP51. The invention also pertains to methods for screening for agents for the prevention and/or treatment of psychiatric disorders, such as depression.
