Methods for controlling sex of oviparous embryos using light sources
10455819 · 2019-10-29 · ·

The sex of embryos in eggs is influenced or controlled through the application of light having selected wavelengths in order to promote the development of embryos of a selected sex. An incubating device is provided having an interior cavity that can be sealed from an outside, and having a plurality of lighting elements disposed on each of a plurality of trays disposed in the interior cavity. Eggs are disposed on the trays, and pre-determined environmental conditions are applied to the interior cavity to promote hatching of the eggs. Concurrently with the application of the environmental conditions, the eggs are irradiated according to pre-determined lighting conditions. The lighting conditions include applying light having wavelengths substantially concentrated in selected ranges, such as light wavelengths within the 390-419 nm, 410-450 nm, 420-450 nm, 450-495 nm, or other narrow range.

Method and apparatus for creating an opening in the calcified shell in the region of the blunt end of an incubated bird egg with embryo and for determining the sex thereof

According to the present invention, a method and an apparatus are provided for creating an opening in the calcified shell of an incubated bird egg containing an embryo, in the region of the blunt end of the incubated bird egg. Within the region of the blunt end there is an outer membrane and an inner membrane with an air cell located therebetween. The apparatus comprises a holder on which the incubated bird eggs are stored with their pointed ends facing downwards, wherein the embryo adjoins the inner membrane, a first detection device configured to detect the position and geometry of the air cell, and an opening device configured to create an opening in the calcified shell at the blunt end of the incubated bird egg above the taut inner membrane. Due to the open calcified shell, the sex of the embryo may be determined in a reliable way.

GENETIC MODIFICATION OF RATS

Compositions and methods are provided for making rat pluripotent and totipotent cells, including rat embryonic stem (ES) cells. Compositions and methods for improving efficiency or frequency of germline transmission of genetic modifications in rats are provided. Such methods and compositions comprise an in vitro culture comprising a feeder cell layer and a population of rat ES cells or a rat ES cell line, wherein the in vitro culture conditions maintain pluripotency of the ES cell and comprises a media having mouse leukemia inhibitory factor (LIF) or an active variant or fragment thereof. Various methods of establishing such rat ES cell lines are further provided. Methods of selecting genetically modified rat ES cells are also provided, along with various methods to generate a transgenic rat from the genetically modified rat ES cells provided herein. Various kits and articles of manufacture are further provided.

Methods for protecting porcine fetuses from infection with Porcine Reproductive and Respiratory Syndrome Virus (PRRSV). The methods comprise breeding a female porcine animal with a male porcine animal. The female porcine animal comprises modified chromosomal sequences in both alleles of its CD163 gene, wherein the modified chromosomal sequences reduce the susceptibility of the female porcine animal to infection by PRRSV, as compared to the susceptibility to infection by PRRSV of a female porcine animal that does not comprise any modified chromosomal sequences in the alleles of its CD163 gene. The male porcine animal comprises at least one wild-type CD163 allele.

ANIMAL MODELS FOR PSORIASIS AND SCREENING METHODS

A biologically relevant animal model for psoriasis is provided. Epidermal-immune interactions governing epidermal tissue homeostasis are altered in psoriasis, an inflammatory disease affecting one in thirty adults. Here, we characterize Rac1 as a key mediator of this process. Rac1 activation was consistently elevated in psoriatic epidermis and primary psoriatic human keratinocytes (PHKC).
