A portable, ex vivo perfusion system for preserving detached biological tissue includes a receptacle for housing the tissue in a normothermic environment, a perfusion core to pump perfusate through the tissue via at least one conduit, at least one detection device to measure parameters during perfusion, and at least one parameter control device to maintain the parameter in a predetermined threshold. The system also include a controller with instructions to receive the measured parameters, compare the parameters to predetermined thresholds, and when the parameters are outside the thresholds change an output of the at least one parameter control device to get the parameters within the threshold and alert a user that parameters were outside the thresholds.

The method for cryopreservation includes treating a substrate with at least one ECM component and/or adding at least one matricellular protein to the substrate or cell culture media, plating the cells on the treated substrate, and cryopreserving the cells. One or more ECM components and/or matricellular proteins that improves cell viability and retention can be identified by evaluating the cells that have been thawed from the cryopreservation temperature to determine cell viability and retention.

Also, provided is a method of obtaining fertilized eggs of marine fish, the method including allowing ovaries, which have been maintained after removal to ovulate and performing artificial insemination. Further, the present invention relates to a method of maintaining the fertilization capability of ovulated eggs of marine fish by using the culture solution. According to the present invention, fertilized eggs of marine fish can be easily obtained and seed can be efficiently produced.
