CONICAL ROTATING SPIN CONTACTOR

A spin contactor is provided for contacting a particulate with a fluid. The spin contactor has a containment section that may contain the particulate, a fluid inlet, and a fluid outlet. The containment section comprises an upper particulate separator, a lower particulate separator, and a conical wall. In most cases, the spin contactor includes a shaft to be operatively coupled to a means for spinning the spin contactor. The fluid is drawn upwardly through the fluid inlet to contact the particulate and then expelled through the fluid outlet. A process of treating a fluid composition is provided by sending the fluid composition through the spin contactor comprising particulates.

A method for monitoring, evaluating and controlling a cyclic chromatographic purification process that involves at least two adsorbers. According to the method, one step is monitoring of the chromatogram, including the measurement of at least one current concentration-proportional signal in the liquid. Another step is conducting an evaluation of the chromatogram, including a comparison of at least one of the current concentration-proportional signals measured in the monitoring step with a threshold value thereof. A further step is controlling the chromatographic purification process by adapting the termination of the currently running phase as a function of the comparison of the evaluation step and initiating the next phase. Finally, according to the method, the sequence of steps is carried out in given order at least twice.

Separation of materials is achieved using affinity binding and acoustophoretic techniques. A column provided with a fluid mixture of materials for separation and support structures may be used with acoustic waves to block flow of the support structures. The support structures can have an affinity for one or more materials in the fluid mixture. By blocking flow of the support structures, materials bound or adhered to the support structure are also blocked.

Perfusion bioreactor with filtration systems

The disclosure provides a filtration system for a cell culture apparatus and a method of cell culture. The filtration system comprises a bioreactor vessel and two or more alternating tangential flow (ATF) filters connected in parallel. A failure in either filter is detected by an in-line sensor, and an automated response system functions to sequester the malfunctioning filter by stopping the flow of liquid media through the filter. Media flow through the remaining operable filters can be increased so that the rate of perfusion through the bioreactor remains relatively unchanged. Such a system may prevent issues that arise from ATF filter failures in conventional perfusion bioreactors, thereby improving the long-term viability of cell cultures.
