The present disclosure provides methods of patterning cells on a surface of a substrate. The methods include disposing a pattern of nucleic acids on a surface of a substrate, and contacting the patterned nucleic acids under hybridization conditions with a first suspension of cells, where cells of the first suspension include cell surface-attached nucleic acids complementary to the patterned nucleic acids, and where the cell surface-attached nucleic acids hybridize to the patterned nucleic acids to pattern the cells on the surface of the substrate. Systems and kits for practicing the methods are also provided.
Arrangement, substrate and method for preparing a cell sample
A flexible, thin, elongated band is used as a substrate. Similarly to a magnetic tape, the band is unwound from a feed reel and is transported past an outlet opening of a receptacle containing the cells such that the cells are poured onto the band. Subsequently, the band containing the cells applied thereon is wound onto a take-up reel. The take-up reel is fixed on a drive shaft which can be rotated by a drive mechanism. The rotation thus achieved has the effect that the band is unwound, transported past the outlet opening and finally wound up. In addition, spacers are provided at the upper surface of the band in order to prevent the contacting of radially adjacent sections of the band containing the cells in a wound-up state.

According to the invention, generally, a method for making a microfluidic aliquoting (MA) chip, adapted to fit in a Petri dish, has a center well (inlet) connected by branched channels to a plurality of side wells (outlets). The chip comes in various types, including a bMA Chip T1, bMA Chip T2, bMA Chip T3, and an rMA Chip. The branched channel improvement provides for a greater distance between neighboring channels and a decreased density near the center well. Design improvements including an injection mold design for an insert and a base and a multiplex hole punch allow for rapid fabrication of the MA chip.
