BISPECIFIC ANTIBODY TREATMENT OF LYMPHOID MALIGNANT NEOPLASM CONDITIONS
Provided is a method for the treatment of a lymphoid malignant neoplasm in a subject in need thereof comprising administering to the subject an effective amount of a bispecific antibody which comprises a Fab portion that binds CD47 and a Fab portion that binds CD20. In some embodiments, the lymphoid malignant neoplasm is Non-Hodgkin's Lymphoma (NHL), follicular lymphoma (FL), diffuse large B-cell lymphoma (DLBCL), marginal zone lymphoma (MZL), mantle cell lymphoma (MCL), or primary mediastinal B-cell lymphoma. In certain embodiments, the lymphoid malignant neoplasm is NHL.
Multispecific antibodies facilitating selective light chain pairing
Provided herein are multispecific antibodies, e.g. bispecific antibodies, which are modified such that the desired chain pairing takes place and/or can be selected for. Specifically, this is achieved by using different dimerization domains for light chain pairing. Also disclosed herein are nucleic acids encoding for these antibodies, expression vectors comprising these nucleic acids, cells expressing them, and further to pharmaceutical compositions comprising the antibodies, as well as methods of isolating the antibodies.
ANTI-LAG3 ANTIBODIES AND ANTIGEN-BINDING FRAGMENTS
The present invention includes antibodies and antigen-binding fragments thereof that specifically bind to human or cynomolgous monkey LAG3 as well as immunoglobulin chains thereof and polynucleotides encoding the same along with injection devices comprising such antibodies or fragments. Vaccines including such antibodies and fragments as well as compositions comprising the antibodies and fragments (e.g., including anti-PD1 antibodies) are included in the invention. Methods for treating or preventing cancer or infection using such compositions are also provided. In addition, methods for recombinant expression of the antibodies and fragments are part of the present invention.
ANTI-PACAP ANTIBODIES AND USES THEREOF
The present invention is directed to antagonistic antibodies and antigen binding fragments thereof having binding specificity for PACAP. These antibodies inhibit, block or neutralize at least one biological effect associated with PACAP, e.g., vasodilation. In exemplary embodiments these antibodies and antigen binding fragments thereof may comprise specific V.sub.H, V.sub.L, and CDR polypeptides described herein. In some embodiments these antibodies and antigen binding fragments thereof bind to and/or compete for binding to specific epitope(s) on human PACAP. The invention is further directed to using these antagonistic anti-PACAP antibodies, and binding fragments thereof, for the diagnosis, assessment, and treatment of diseases and disorders associated with PACAP and conditions where antagonism of PACAP-related activities, such as vasodilation, mast cell degranulation, and/or neuronal activation, are therapeutically beneficial, e.g., headache and migraine indications.

Ultralong complementarity determining regions and uses thereof
Disclosed herein are immunoglobulin constructs comprising at least one immunoglobulin domain or fragment thereof; and a therapeutic polypeptide or derivative or variant thereof attached to or inserted into said immunoglobulin domain. Also provided are immunoglobulin constructs comprising a mammalian immunoglobulin heavy chain comprising at least a portion of a knob domain in the complementarity-determining region 3 (CDR3H) or fragment thereof; and a therapeutic polypeptide attached to or inserted into said knob domain of the CDR3H. Also provided are immunoglobulin constructs comprising a mammalian immunoglobulin heavy chain comprising at least a portion of a stalk domain in the complementarity-determining region 3 (CDR3H) or fragment thereof; and a therapeutic polypeptide attached to or inserted into said stalk domain of the CDR3H. Activatable Anti-CTLA-4 Antibodies and Uses Thereof
Provided herein are activatable anti-human CTLA-4 antibodies comprising a heavy chain comprising a VH domain and a light chain comprising a masking moiety (MM), a cleavable moiety (CM), and a VL domain. Such activatable anti-human CTLA-4 antibodies have CTLA-4 binding activity in the tumor microenvironment, where the masking moiety is removed by proteolytic cleavage of the cleavable moiety by tumor-specific proteases, but exhibit greatly reduced binding to CTLA-4 outside the tumor. In this way, the activatable anti-human CTLA-4 antibodies of the present invention retain anti-tumor activity while reducing the side effects associated with anti-CTLA-4 activity outside the tumor.
