A bioreactor includes a base and a lid. The base includes two opposing curved or convoluted surfaces, two opposing flat surfaces, a window disposed on one of the flat surfaces, the window having a higher degree of transparency compared to other portions of the base, wherein images or videos of cells are captured through the window by non-invasive ISM device, and a rounded bottom. The lid is attachable to the base. The lid includes a shaft and a semipermeable membrane attached to the shaft, the semipermeable membrane being permeable to oxygen but impermeable to viruses and bacteria. The PAT-based online analysis directs the adaptive manipulations of bioreactor towards efficient, automated, and GMP-compliant clinical protocols of cell culture.

CULTURE METHOD

Provided is a culture method for a microorganism using a plate-like microchamber having a plurality of wells independent from each other on a front surface thereof. The culture method for the microorganism includes: mixing a liquid culture medium containing agarose having a gelation temperature of no less than 5° C. and no more than 20° C. and the microorganism to obtain a mixture; enclosing the mixture into the well with the front surface facing upward; and keeping the microchamber in which the mixture is enclosed in the well at a predetermined temperature range.

A process conducted in a rocking bioreactor is monitored using a flexible bag fitted with an in-situ probe. The probe includes a disposable bag insert or patch and a reusable probe module. The patch is secured to a wall of the bag, and the probe module is a detachable part that can be mated to the bag patch, then disassembled for future use, for example. The patch is secured to the bag, defining a sample gap, and the probe module is inserted or mated with the patch. The probe module includes one or more source elements and one or more detector element. Light is transmitted from the source elements, through the sample gap, to the detector element(s), which detect the light after interaction with contents of the bag.
