A microporous substrate for detection of surface bound target analyte molecules includes a microporous substrate material having opposed surfaces and tapered micropores extending through the substrate with the micropores having wider openings on one side of the substrate compared to the other side. The micropores have bound therein analyte specific receptors complementary to the target molecules. When a liquid sample containing the target analyte molecules with optical probes attached to the target molecules is flowed through the substrate, they bind to their complementary analyte specific receptors and emit light. This microporous substrate structure gives an increase in the collection efficiency of light emitted from optical probes when the light is detected by a light detector spaced from the side of the microporous substrate facing the larger micropores openings compared to a light collection efficiency of light emitted from the optical probes when the micropores are straight and not tapered.

SYSTEMS AND METHODS PRODUCING SEEDED GRAFTS

Closed disposable seeding systems with improved seeding chambers permitting uniform seeding of a scaffold or graft with patient's cells are provided. The seeding chambers with a variable width along the length of the chamber, or a minimal gap between the scaffold and chamber wall, provide an improvement of the prior seeding chambers of closed disposable seeding systems by providing faster and more efficient and uniform seeding of the grafts and scaffolds. Also described are scaffolds with biomechanical and structural properties permitting spontaneous reversal of stenosis and neotissue formation as the graft degrades yielding a scaffold-free neovessel.

Self-assembling multicellular bodies and methods of producing a three-dimensional biological structure using the same

Structures and methods for tissue engineering include a multicellular body including a plurality of living cells. A plurality of multicellular bodies can be arranged in a pattern and allowed to fuse to form an engineered tissue. The arrangement can include filler bodies including a biocompatible material that resists migration and ingrowth of cells from the multicellular bodies and that is resistant to adherence of cells to it. Three-dimensional constructs can be assembled by printing or otherwise stacking the multicellular bodies and filler bodies such that there is direct contact between adjoining multicellular bodies, suitably along a contact area that has a substantial length. The direct contact between the multicellular bodies promotes efficient and reliable fusion. The increased contact area between adjoining multicellular bodies also promotes efficient and reliable fusion. Methods of producing multicellular bodies having characteristics that facilitate assembly of the three-dimensional constructs are also provided.

A bioreactor for culturing of cells is described. Screens suitable as a cell growth scaffold may comprise crossed fibers. Screens may be contained loosely in a screen holder, which in turn may be contained inside a manifold assembly. A lower manifold, screen holder and upper manifold may have identical or similar interior open cross-sections. Flow of liquid medium can occur upwardly through the array of screens, then flowing over a weir in the presence of an air pocket, and into a moat and a pump. The screen holder may have slots whose exterior-facing ligaments are rounded, and may have grooves whose interior-facing edges are rounded. These components may be located inside an incubator suitable to maintain desired environmental conditions and cleanliness.

Drug screening platform simulating hyperthermic intraperitoneal chemotherapy

A drug screening platform simulating hyperthermic intraperitoneal chemotherapy including a dielectrophoresis system, a microfluidic chip and a heating system is disclosed. The dielectrophoresis system is used to provide a dielectrophoresis force. The microfluidic chip includes a cell culture array and observation module and a drug mixing module. The cell culture array and observation module are used to arrange the cells into a three-dimensional structure through the dielectrophoresis force to construct a three-dimensional tumor microenvironment. The drug mixing module is coupled to the cell culture array and observation module and used to automatically split and mix the inputted drugs and output the drug combinations into the cell culture array and observation module. The heating system is used for real-time temperature sensing and heating control of the drug combinations on the microfluidic chip to simulate high-temperature drug environment when performing hyperthermic intraperitoneal chemotherapy on the three-dimensional tumor microenvironment.
