FIXED-BED BIOREACTOR WITH CONSTANT-FLOW PUMP/ TUBING SYSTEM

We have modified a commercially-available adherent cell culture bioreactor in several ways to increase productivity of cultured cells, while decreasing contamination risk. We found that modifying a commercially-available adherent cell culture bioreactor to provide for slower cell culture medium flow unexpectedly and dramatically increases the productivity of the cultured adherent cells. We also developed a new sampling manifold configuration and new way of taking samples, to reduce contamination risk.

The present disclosure provides a perfusion system and a cell cultivation perfusion system, the perfusion system comprising: a pressure transfer component, the pressure transfer component including a belt and multiple pressure members disposed on the belt at intervals; a driving device, a driving connection being established between the driving device and the pressure transfer component; and at least one pipeline assembly, the at least one pipeline assembly including at least one pressable pipeline for fluid flow; wherein a press area is formed between the pressure transfer component and each of the at least one pressable pipeline, the multiple pressure members move with the belt, and at least a portion of the multiple pressure members press against the at least one pressable pipeline in the press area.

WOVEN CELL CULTURE SUBSTRATES, BIOREACTOR SYSTEMS USING THE SAME, AND RELATED METHODS

A cell culture matrix for a perfusion-flow fixed-bed reactor is provided. The cell culture matrix includes a substrate having a porous sheet for adhering cells thereto. The sheet has a first side, a second side opposite the first side, a thickness separating the first side and the second side, and a plurality of openings formed in the substrate, arrayed in a regular pattern, and passing through the thickness of the substrate. The porous sheet is wound into a cylindrical shape having a plurality of wound layers, and the cell culture matrix does not include a spacer material or physical barrier between the plurality of wound layers of the substrate.

APPARATUSES AND SYSTEMS FOR PREPARING A MEAT PRODUCT

The apparatuses described herein relate to preparation of a meat product. Apparatuses, systems comprising the apparatuses, and methods of making and use the systems and apparatuses are described herein. These are useful for controlling one or more of growth on and separation of a meat product from an enclosed substrate. The apparatuses and systems are configured to receive fluid and grow the meat product and/or separate the meat product from the substrate in a scalable manner.

Some aspects of this disclosure provide methods of producing decellularized and recellularized cardiac tissues engineering methods of treating heart disease, including heart aneurysm and myocardial infarction. Cardiac tissue engineering of compositing and constructing of a recellularized coronary-based right heart flap or cardiac flap scaffold with its cellular components are also disclosed.

Expanding cells in a bioreactor

Described are embodiments for expanding cells in a bioreactor. In one embodiment, methods are provided that distribute cells throughout the bioreactor and attach cells to specific portions of a bioreactor to improve the expansion of the cells in the bioreactor. Embodiments may be implemented on a cell expansion system configured to load, distribute, attach and expand cells.

A bioprocess vessel includes a flexible bag or substantially rigid container that defines an interior volume and having a bottom surface, the bottom surface being open or containing an aperture therein for the passage of fluid. A pump is secured to the bottom surface of the flexible bag or substantially rigid container. In some embodiments is secured directly to the flexible bag or substantially rigid container. In other embodiments, the pump is secured indirectly the flexible bag or substantially rigid container using, for example, a port that extends through the aperture on the bottom surface. The port or flanged surface may also be integrated into the pump, which is secured to the vessel. An optional mixing adaptor may be provided inside the interior volume of the flexible bag or substantially rigid container and at least partially covers the inlet that leads to the pump.
