APPARATUS FOR EFFICIENT GENETIC MODIFICATION OF CELLS

A device for treatment of cells with particles is disclosed. The device includes a semi-permeable membrane positioned between two plates, the first plate defining a first flow chamber and comprising a port, a flow channel, a transverse port, and a transverse flow channel, the first flow chamber constructed and arranged to deliver fluid in a transverse direction along the first side of the semi-permeable membrane, the second plate defining a second flow chamber and comprising a port. A method for transducing cells is disclosed. The method includes introducing a fluid with cells and viral particles into a flow chamber adjacent a semi-permeable membrane such that the cells and the viral particles are substantially evenly distributed on the semi-permeable membrane. The method also includes introducing a recovery fluid to suspend the cells and the viral particles, and separating the cells from the viral particles. A method of activating cells is disclosed.

A perfusion module and a perfusion culture system are provided, including: a bioreaction module, including at least one bioreactor; a perfusion module, including at least one perfusion chamber, each perfusion chamber is connected to a pressure regulating pipe for inflow or outflow of an airflow; when there are multiple perfusion chambers, every two adjacent perfusion chambers are connected to each other; at least one bioreactor, the perfusion module, the filtration module are communicated in sequence, if there are more than two perfusion chambers, all of perfusion chambers are set side by side; a cell culture fluid in at least one bioreactor flows into at least one perfusion chamber, flows out from at least one perfusion chamber into at least one filter, and a permeate from at least one filter flows into a harvesting device. The present disclosure avoids high shear forces in the perfusion culture system.

Embodiments described herein generally provide for expanding cells in a cell expansion system. The cells may be grown in a bioreactor, and the cells may be activated by an activator (e.g., a soluble activator complex). Nutrient and gas exchange capabilities of a closed, automated cell expansion system may allow cells to be seeded at reduced cell seeding densities, for example. Parameters of the cell growth environment may be manipulated to load the cells into a particular position in the bioreactor for the efficient exchange of nutrients and gases. System parameters may be adjusted to shear any cell colonies that may form during the expansion phase. Metabolic concentrations may be controlled to improve cell growth and viability. Cell residence in the bioreactor may be controlled. In embodiments, the cells may include T cells. In further embodiments, the cells may include T cell subpopulations, including regulatory T cells (Tregs), for example.

A storage assembly for storing a plurality of specimens includes a frame, a plurality of modular storage units for perfusion and/or incubation of one or more specimens removably coupled to the frame, a sample transfer apparatus configured to retrieve a specimen holder from a chosen modular storage unit of the plurality of modular storage units, and a control unit communicatively coupled to the sample transfer apparatus. The control unit is configured to cause the sample transfer apparatus to retrieve a specimen from a modular storage unit of the plurality of modular storage units and deliver the specimen to a delivery position.

The apparatuses described herein relate to preparation of a meat product. Apparatuses, systems comprising the apparatuses, and methods of making and use the systems and apparatuses are described herein. These are useful for controlling one or more of growth on and separation of a meat product from an enclosed substrate. The apparatuses and systems are configured to receive fluid and grow the meat product and/or separate the meat product from the substrate in a scalable manner.

MIMICKING PHYSIOLOGIC CONDITIONS DURING ORGAN OR TISSUE PERFUSION

A system is configured to perfuse one or more organs and/or associated tissues while monitoring the physiological function, testing medical devices or therapies, or both. The system includes a pump, a container, a fluid circuit, a platform, and at least one actuator. The pump is configured to generate a flow of a fluid. The container defines an interior cavity and is configured to receive at least one organ or tissue and maintain at least one environmental condition associated with the at least one organ or tissue within the interior cavity. The fluid circuit is configured to fluidically couple the pump to the at least one organ or tissue. The platform is operably coupled to the container. The at least one actuator is configured to move the platform to mimic at least one biological movement associated with the at least one organ or tissue.

Fluidic devices including microplates with interconnected wells

A fluidic device for culturing cells includes a microplate and plate lid. The microplate includes multiple wells and channels, the channels extending between the wells such that the channels interconnect the wells. The plate lid releasably engages the microplate to thereby enclose the wells and the channels. The wells include a culture surface such that a cell culture medium received therein is deposited over the culture surface. At least one channel that extends between adjacent ones of the wells is spaced from the culture surfaces of the adjacent wells defining a gap between the at least one channel and the culture surfaces of the adjacent wells for collection of the cell culture medium.
