Woven cell culture substrates, bioreactor systems using the same, and related methods
A cell culture matrix for a perfusion-flow fixed-bed reactor is provided. The cell culture matrix includes a substrate having a porous sheet for adhering cells thereto. The sheet has a first side, a second side opposite the first side, a thickness separating the first side and the second side, and a plurality of openings formed in the substrate, arrayed in a regular pattern, and passing through the thickness of the substrate. The porous sheet is wound into a cylindrical shape having a plurality of wound layers, and the cell culture matrix does not include a spacer material or physical barrier between the plurality of wound layers of the substrate.
Integrated continuous isolation of fluid streams from sterile process vessels
Provided herein are isolation processes and the associated hardware to allow fluid streams to be isolated from a sterilized system (e.g., a sterile process vessel) that contains a sterile process. The isolation processes described herein allow for continuous removal of fluid streams (e.g., waste streams, liquid containing recombinant therapeutic proteins) from a sterilized system (e.g., a biological manufacturing system), which provides for less manual manipulation of the sterilized system and a decreased risk of contaminating the sterilized system.
CULTURE VESSEL, CULTURING SYSTEM, AND METHOD FOR PRODUCING CULTURE
According to one embodiment, a culture vessel includes a rigid bottom part and a wall part. The wall part has a culture medium inlet and includes a deformable portion. The culture vessel allows the deformable portion to deform so as to change the capacity of the culture vessel when a pressure of the internal space of the culture vessel varies.
Aseptic fluid couplings
Some fluid coupling devices described herein are configured for use in fluid systems. For example, some embodiments described in this document are single-use, aseptic fluid coupling devices that can be coupled to create a sterile flow path therethrough. Some such aseptic couplings are genderless couplings such that two identical aseptic couplings can be coupled together and then latched to form a robust connection between the aseptic couplings.
FIXED BED BIOREACTOR AND METHODS OF USING THE SAME
A fixed-bed bioreactor system is provided that includes a vessel with a media inlet, a media outlet, and an interior cavity disposed between and in fluid communication with the media inlet and media outlet. The vessel further includes a cell culture substrate disposed in the interior cavity between the media inlet and the media outlet in a packed-bed configuration, the cell culture substrate including a plurality of porous disks in a stacked arrangement. The interior cavity includes a cell culture section and a spacer section, the cell culture substrate defining the cell culture section and the spacer section being disposed between the cell culture section and the media outlet, and each of the plurality of porous disks has a surface configured to culture cells thereon.
AN APPARATUS FOR THE INCUBATION OF A BIOLOGICAL MATERIAL
The invention relates to an apparatus (200) for incubation of a viable biological material (2); said apparatus comprises: a housing (4) having an extension in a longitudinal direction X, in a transversal direction Y, and in a direction Z perpendicular to the longitudinal direction and the transversal direction; said housing comprising: two or more culture dish compartments (6), each being adapted to accommodate, one or more culture dishes (8) comprising a biological material (2); wherein said apparatus comprises an image capturing device (10); wherein said apparatus comprises a control unit (12) for controlling the operation thereof; wherein at least part of said image capturing device is being configured to be movable in relation to the two or more culture dish compartments (6), thereby allowing capture of images of one or more of said biological materials (2) accommodated in said one or more culture dishes (8); and wherein said apparatus comprises a FLIM unit (11) (fluorescent lifetime imaging microscope); wherein at least part of said FLIM unit (11) is being configured to be movable in relation to the two or more culture dish compartments (6), thereby allowing capture of FLIM spectra of one or more of said biological materials (2) accommodated in said one or more culture dishes (8).
AEROBIC FERMENTATION SYSTEMS AND METHODS
A system for aerobic fermentation includes a vessel, an aeration system including a gas sparger fluidly coupled to the vessel to introduce a compressed gas to an internal volume of the vessel, and a recirculation loop fluidly coupled to an outlet of the vessel. The recirculation loop includes an eductor fluidly coupled to an oxygen-containing gas source, a static mixer downstream of the eductor, a heat exchanger downstream of the eductor, and a distributor downstream of the heat exchanger. The distributor is fluidly coupled to the vessel. The aeration system provides mixing and oxygen mass transfer to the fermentation composition in the vessel. The fermentation composition passes through the eductor, static mixer, heat exchanger, and distributor of the recirculation loop, and back into the vessel. Oxygen is transferred from an oxygen containing gas to the fermentation composition and heat is removed from the fermentation composition in the recirculation loop.
Midscale Model For Organic Growth and Phasing
The present disclosure is directed to the organic growth and phasing design for a system for the industrial growth of biologics. A system may include a number of subsystems, such as a buffer distribution subsystem, a media preparation subsystem, a bioreactor subsystem, a harvest subsystem, and/or a purification subsystem. The subsystems may be highly interconnected for flexible process flow design. Additionally, the subsystems may be constructed with a total output capacity with a total number of equipment stations and operated at a lower output capacity with fewer than the total equipment stations to maintain headroom for growth in phases.
