A diagnostic test system, including: a diagnostic test assembly and a diagnostic test apparatus to perform a test on a biological or environmental sample; the diagnostic test assembly includes: a sample preparation reservoir to receive the sample into a sample preparation fluid, such that a swab carrying the sample can be used to stir the preparation fluid and to wash the swab; a sample dispensing mechanism for insertion into the sample preparation reservoir; a closure to seal the sample preparation reservoir; at least one diagnostic test reservoir coupled to the sample preparation reservoir; and at least one seal between the sample preparation reservoir and the diagnostic test reservoir to prevent fluid movement between the respective reservoirs; wherein the sample dispensing mechanism is operable to disrupt the seal to allow sample fluid to enter the diagnostic test reservoir from the sample preparation reservoir, and to dispense a predetermined amount of fluid.

Large scale processes for producing high purity samples of biologically active molecules of interest from bacterial cells are disclosed. The methods comprise the steps of producing a lysate solution by contacting a cell suspension of said plurality of cells with lysis solution; neutralizing said lysate solution with a neutralizing solution to produce a dispersion that comprises neutralized lysate solution and debris; filtering the dispersion through at least one filter; performing ion exchange separation on said neutralized lysate solution to produce an ion exchange eluate; and performing hydrophobic interaction separation on said ion exchange eluate to produce a hydrophobic interaction solution. Further, provided are compositions comprising large scale amounts of plasmid DNA produced by the disclosed large scale processes.

The object is to provide a lysis method, lysis treatment solution, detection method using an immunochromatographic device, and detection kit comprising an immunochromatographic device for detecting whether causative bacteria of mastitis are coliform bacteria or not by using milk of a livestock animal. There is provided a method for lysing coliform bacteria, which comprises the step of mixing a lysis agent containing a lytic enzyme, and at least one kind of anionic surfactant, and preferably further containing at least one kind of nonionic surfactant, with milk obtained form a livestock animal to lyse coliform bacteria existing in the milk. The lytic enzyme is preferably lysozyme.

Provided herein is a method to reduce the endotoxin contamination in plasmid preparations. In the described method, plasmid DNA and endotoxins are bound to an anionic exchange matrix and are brought into contact with a wash buffer, comprising a combination of branched secondary alcohol ethoxylates with varying ethylene oxide chain lengths, wherein the branched secondary alcohol ethoxylates with the shorter ethoxylate chain is present in the washing buffer in excess compared to the branched secondary alcohol ethoxylate with the longer ethoxylate chain. The resulting purified plasmid has minimal endotoxin contamination levels and is considered endotoxin-free. Furthermore, provided are wash buffers comprising a combination of branched secondary alcohol ethoxylates with varying ethylene oxide chain lengths, kits comprising such wash buffers and the use of such wash buffers for reducing the endotoxin contamination in plasmid preparations.
