TREATMENT OF PARKINSON'S DISEASE AND OTHER CONDITIONS CAUSED OR MEDIATED BY SENESCENT ASTROCYTES USING SMALL MOLECULE SENOLYTIC AGENTS

This invention establishes a new paradigm for treatment of Parkinson's disease (PD) by eliminating senescent cells that reside in or around the site of the disease pathophysiology. Exposure of test subjects to the herbicide paraquat (PQ) increases the risk for developing Parkinson's disease. The data in this disclosure show that PQ induces a senescence arrest and SASP in astrocytes, in culture and in vivo in mice, and senescent cell markers were present in astrocytes in midbrain tissue from PD patients. In a transgenic mouse model, senescent cell ablation protected against PQ-induced PD-like neuropathology. Removal of senescent cells from affected sites using small molecule agents that specifically target senescent cells can help prevent or ameliorate signs and symptoms of the disease.

Therapeutic serum obtained from co-cultured cells

A therapeutic serum suitable for inclusion in a cosmetic preparation may be produced by stressing a co-culture including proliferative cells. The co-culture of cells may be obtained by growing first culture to less than one-hundred percent confluence on a surface. After a monolayer of first culture is established, a second culture may be seeded onto at least one cell free area on the surface, the resulting co-culture grown to less than one-hundred percent confluence. Additional cultures may then be seeded onto cell free areas of the surface and established until a monolayer having the desired population of cells is obtained. The monolayer is then stressed to obtain a serum by conditioning a collection medium. The obtained serum may be combined with a suitable cosmetic base to provide a cosmetic preparation.

METHODS FOR REGULATING POTENCY OF PLURIPOTENT STEM CELLS AND APPLICATIONS THEREOF

The present invention relates to a method for regulating potency of pluripotent stem cells (PSCs) by modulating expression of podocalyxin-like protein 1 (PODXL) and applications thereof.

Systems and Methods for Processing Tissue

Embodiments of the present specification provide systems and methods for holding one or more tissues, such as veins, such that the tissue remains open in a chamber while undergoing orbital shaking with various solutions and to allow uniform treatment during a decellularization process. A frame is held on a stand to which the tissues are attached and comprises a tension inducing mechanism to cause the tissues to controllably stretch. The frame is removed from the stand, with the tissue attached to it, and placed in a decellularization chamber for uniform treatment of the tissue.

SHORT HAIRPIN RNA (SHRNA734) AND USE OF SAME TO POSITIVELY SELECT AND ELIMINATE GENETICALLY MODIFIED CELLS

A potent short hairpin RNA (shRNA734) directed to human Hypoxanthine Guanine Phosphoribosyltransferase (HPRT) improves the rate of gene-modified stem cell engraftment by a conditioning and in vivo selection strategy to confer resistance to a clinically available guanine analog antimetabolite, 6TG, for efficient positive selection of gene-modified stem cells. Uses for polynucleotides comprising the shRNA734 include methods for knocking down HPRT in a cell, for conferring resistance to a guanine analog antimetabolite in a cell, for producing selectable genetically modified cells, for selecting cells genetically modified with a gene of interest from a plurality of cells, for removing cells genetically modified with a gene of interest from a plurality of cells, and for treating a subject infected with HIV.
