SYSTEM AND METHOD FOR AUTOMATIC NUCLEIC ACID EXTRACTION AND QUIALITATIVE ANALYSIS
The present invention provides a system and method for automatic nucleic acid extraction and qualitative analysis. The system comprises a magnetic rotary mixer which comprises a plurality of magnetic rods for generating magnetism, configured to be retractable from the magnetic rotary mixer; a plurality of spin shaft for mounting tips, and the plurality of magnetic rods extend therein; an auto stage comprises a plate holder, which allows a plate place thereon; a mixer holder to hold the magnetic rotary mixer over the plate holder; and a heat plate, disposed under the plate holder for heating the plate. The present invention provides an automated high-throughput nucleic acid extraction and qualitative diagnosis with high efficiency and high accuracy, which is easy to interpret for operators, and realize that nucleic acid extraction and molecular detection can be completed at one time in a single device.
UNIVERSAL LAMP ASSAYS FOR DETECTION OF NUCLEIC ACID TARGETS
Disclosed are compositions and methods that enable loop-mediated isothermal amplification (LAMP) of one or more nucleic acid targets without the need for conventional LAMP primer design customized to each target. A transduction reaction is performed upstream from the LAMP reaction. The transduction reaction generates a single stranded DNA (ssDNA) oligonucleotide when the target nucleic acid is present in the sample. The ssDNA generated in the transduction reaction functions as a required LAMP primer for a universal LAMP template. The ssDNA thus promotes the LAMP reaction. Analysis of the LAMP products can determine the presence of the one or more nucleic acid targets.

Methods for Detecting Low Levels of Covid-19 Virus
Provided herein is a method for detecting the presence of a COVID-19 virus in a human sample or an environmental sample having one or more viruses and bacterial pathogens. Samples are processed to obtain total nucleic acids. A combined reverse transcription and asymmetric PCR amplification reaction is performed to obtain fluorescent labeled COVID-19 virus specific amplicons. The amplicons are detected by microarray hybridization near the lowest limit of detection. Also provided is a method for detecting concurrently with COVID-19 virus, the presence of respiratory disease-causing pathogens including viruses, bacteria and fungus in a single assay using the above method.
