Methods for using tails and tail tags to perform sequencing of nucleic acid molecules are also disclosed. Tails and tail tags are constructs associated with nucleic acid molecules based on their nucleotide base composition. In many embodiments, a removable tail is associated with a nucleotide comprising a specific base type and incorporated into a nucleic acid molecule. The removable tail facilitates attachment of a tail tag to the nucleic acid molecule, said tail tag representing the base type of said nucleotide. Removal of the removable tail and repetition of the process generates a series of attached tail tags that represent the sequence of the nucleic acid molecule. The series of attached tail tags can be readily detected by nanopore devices, thus revealing the sequence of the nucleic acid molecule.
