Processes and systems related to solid phase peptide synthesis are described. The processes include a flow-through process comprising cyclic addition of amino acids to a column packed with resin, wherein each cycle includes the combination of the amino acids with one or more reagents to provide an activated amino acid mixture, and wherein heating is applied to the amino acid(s) before they are passed through the column and wherein the amino acids are re-circulated at least once over the column packed with resin. Systems for such processes are also described.

An improvement in of deprotection in solid phase peptide synthesis is disclosed. The method includes the steps of adding the deprotection composition in high concentration and small volume to the mixture of the coupling solution, the growing peptide chain, and any excess activated amino acid from the preceding coupling cycle; and without any draining step between the coupling step of the previous cycle and the addition of the deprotection composition for the successive cycle; and with the coupling solution at a temperature of at least 30° C.

An improved method for coupling amino acids into peptides or peptidomimetics is disclosed that includes the steps of combining an amino acid, a carbodiimide, an activator additive, and a base at less than 1 equivalent compared to the amino acid to be activated; and carrying out the activation and coupling at a temperature greater than 30° C.

SOLID PHASE PEPTIDE SYNTHESIS METHODS AND ASSOCIATED SYSTEMS

Methods and system for solid phase peptide synthesis are provided. Solid phase peptide synthesis is a known process in which amino acid residues are added to peptides that have been immobilized on a solid support. New amino acid residues are added via a coupling reaction between an activated amino acid and an amino acid residue of the immobilized peptide. Amino acids may be activated using, e.g., a base and an activating agent. Certain inventive concepts, described herein, relate to methods and systems for the activation of amino acids. These systems and methods may allow for fewer side reactions and a higher yield compared to conventional activation techniques as well as the customization of the coupling reaction on a residue-by-residue basis without the need for costly and/or complex processes.

An improved method of deprotection in solid phase peptide synthesis is disclosed. In particular the deprotecting composition is added in high concentration and small volume to the mixture of the coupling solution, the growing peptide chain, and any excess activated acid from the preceding coupling cycle, and without any draining step between the coupling step of the previous cycle and the addition of the deprotection composition for the successive cycle. Thereafter, the ambient pressure in the vessel is reduced with a vacuum pull to remove the deprotecting composition without any draining step and without otherwise adversely affecting the remaining materials in the vessel or causing problems in subsequent steps in the SPPS cycle.

Method of preparing stapled peptides

Described herein is an operationally simple, one-pot solid-supported preparation of saturated stapled peptides. Following completion of ruthenium-catalysed metathesis, solid-phase transfer hydrogenation was achieved using triethylhydrosilane at elevated temperatures. The utility of the method has been demonstrated on 14- and 16-mer peptides to yield the corresponding cyclic a-helix stabilised stapled peptides.

A method and apparatus for nucleic acid synthesis. The method employs a device including at least one deprotection unit to carry out a step of deprotection, at least one coupling unit to carry out a step of coupling, at least one oxidation/thiolation unit to carry out a step of oxidation orthiolation, at least one capping unit to carry out a step of capping, and at least one washing unit to carry out a step of washing. A plurality of reaction vessels for nucleic acid synthesis are moved to the units in accord with a synthesis scheme for a desired nucleic acid sequence and at least two reaction vessels are simultaneously acted upon at several of the units in series.

TREATMENT APPARATUS

In order to provide a treatment apparatus that can efficiently perform microwave irradiation, a treatment apparatus includes: a vessel made of a microwave-reflecting material, and having a first end and an irradiation opening portion, which is an emitting portion of microwaves that are emitted into the vessel; a first filter located so as to partition the vessel, and configured to separate solids that are to be separated, from the contents of the vessel; and a first reflecting member located closer to the first end than the emitting portion is and so as to partition the vessel, and configured to allow at least the contents having passed through the first filter to pass through the first reflecting member, and to reflect microwaves.

A continuous flow reactor, a method of performing a continuous flow reaction, and a method of controlling a moveable wall of a reaction chamber of a continuous flow reactor. The reactor comprising: an inlet; an outlet; and a reaction chamber, between the inlet and the outlet and providing a flow path therebetween, the reaction chamber having a moveable wall; the reactor further comprising: a pressure sensor configured to monitor a fluid pressure in the continuous flow reactor; and a controller, operable to adjust the position of the moveable wall, and thereby change a volume of the reaction chamber, based on the monitored fluid pressure.
