MULTISPECIFIC POLYPEPTIDE CONSTRUCTS CONTAINING A CONSTRAINED CD3 BINDING DOMAIN AND A RECEPTOR BINDING REGION AND METHODS OF USING THE SAME
The invention relates generally to multispecific polypeptides that bind at least CD3, a second antigen, and a receptor of a T cell, such as a costimulatory receptor or an inhibitory receptor, in which the multispecific polypeptide constructs are able to engage CD3. In some embodiments, the multispecific polypeptide constructs bind a costimulatory receptor and provide costimulatory binding activity. In some embodiments, the multispecific polypeptide constructs bind an inhibitory receptor and block inhibitory activity. In some aspects, the multispecific polypeptides have constrained CD3 binding and bind to or engage CD3 only upon binding to the second antigen, such as a tumor associated antigen. In some embodiments, the multispecific polypeptides contain cleavable linkers that, when cleaved, result in dual effector functions. Also provided are methods of making and using these multispecific polypeptides in a variety of therapeutic, diagnostic and prophylactic indications.
In vivo method for generating diversity in a protein scaffold
A transgenic non-human animal is provided. In certain embodiments, the animal comprises a genome comprising an immunoglobulin heavy chain locus comprising: a) a transcribed gene encoding a fusion protein comprising, from N-terminus to C-terminus: i. a scaffold comprising a first binding domain; and ii. a heavy chain constant region operably linked to the scaffold; wherein the scaffold is capable of specifically binding to a target in the absence of additional polypeptides; and b) a plurality of pseudogenes that are operably linked to the transcribed gene and that donate, by gene conversion, nucleotide sequence to the part of the transcribed gene that encodes the binding domain.

There are many reports of diseases caused by overeating, satiation, and an unbalanced diet, and various therapeutic methods and therapeutic drugs are proposed for said diseases. There have not been many proposals, for prevention of these diseases, of methods by which what is eaten is not readily absorbed in vivo. The present invention involves inoculating female birds with a digestive enzyme as an antigen, said digestive enzyme being present in vivo. As a result of inhibiting the activity of the digestive enzyme by using an antibody which has been produced in vivo in the birds, the present invention inhibits decomposition of proteins, lipids, and carbohydrates and reduces in vivo absorption. This type of antibody can be obtained from an egg laid by a female bird which has received the antigen, and furthermore the egg itself includes the antibody. Consequently, a food product that contains, as an ingredient thereof, eggs having this type of antibody is low in proteins, lipids, and carbohydrates. Furthermore, by ingesting the antibody obtained from the eggs, the absorption of proteins, lipids, and carbohydrates can be decreased.

ANTIBODY AND ANTIBODY-CONTAINING COMPOSITION
For many diseases due to microbes or the like, proliferation of microbes themselves is a cause of a symptom. However, there were cases where a substance released by the microbes is a cause of a symptom. In such cases, when attempting to treat a disease with an antibody, it was necessary to obtain an antibody against an antigen that is a substance causing the disease. However, it was difficult to find the underlying substance causing the disease among substances released by the microbes. An antibody (polyclonal) binding to not only an antigen but also to a substance, which is secreted by the antigen and accelerates the deterioration of a symptom, is obtained by immunizing birds with a lysis solution produced from lysing microbial cells as an antigen. Further, an antibody obtained with a surface protein of a virus as an antigen is expected to inhibit an infection by a virus.
BINDING AGENTS
The invention relates to antibody molecules and antigen-binding portions thereof which bind specifically to glucocorticoid-induced TNF receptor (GITR). In particular aspects of the invention, the antibody molecules specifically bind to human GITR and cynomolgus monkey GITR. The anti-GITR antibody molecules of the invention have been developed and optimized using CDR sequences derived from a murine anti-GITR antibody 6C8. Medical uses of the antibody molecules are disclosed.
