The present technology provides a cell treatment system including a cell culture unit capable of culturing a cell group, a measurement unit capable of measuring the cell group, and an estimation unit, in which the estimation unit estimates, on the basis of information regarding a cell before treatment of the cell group and at least one of information regarding a predetermined cell to reach after treatment of the cell group or a predetermined treatment condition for the cell group before treatment, a treatment condition under which the cell group is derivable to the information regarding the predetermined cell or information regarding the cell after treatment of the cell group derived by the predetermined treatment condition.
UNIFORM CELL CULTURE SUBSTRATE FOR FIXED BED BIOREACTOR
A bioreactor system for culturing cells is provided having a cell culture vessel with an interior reservoir, and an inlet and outlet connected to the reservoir. The system also includes a cell culture matrix in the reservoir for adhering cells thereto. The cell culture matrix maintains a uniform flow of liquid media through the cell culture matrix, such that the uniform flow satisfies the following expressions: wherein P is greater than or equal to at least one of 0.3, 0.4, 0.5, 0.6, 0.7, 0.8, 0.9, and 0.95, and wherein N is less than or equal to at least one of 0.1, 0.2, 0.3, 0.4, 0.5, 0.6, 0.7, 0.8, 0.9, and 1.0.
BIOREACTOR SYSTEM WITH EXTERNAL LOOP FOR ENHANCED GAS TRANSFER AND/OR TRANSFECTION
An apparatus is provided for culturing and transfecting cells using a liquid. A bioreactor includes a cell culture bed for receiving the liquid. An external loop is connected to the bioreactor and adapted for circulating the liquid to and from the bioreactor. The external loop is adapted for introducing a gas into the liquid therein and/or for circulating a transfection material in the liquid therein to the bioreactor. In one embodiment, a first vessel is associated with the external loop for receiving the liquid, and a gas injector is provided for injecting a gas into the liquid, such as into the first vessel. A second vessel is also provided for providing a transfection material (such as a transfecting agent as part of a transfection mixture) to the liquid, such as directly to the bioreactor or to the external loop. In another embodiment, the gas injector is associated directly with the external loop. The external loop may be dedicated for gas injection and/or transfection, and maintained separate from a loop for circulating liquid media to the bioreactor, which potentially allows for operation on demand. Related methods are also disclosed.
CULTURE DEVICE FOR TISSUE CELL SUSPENSION
The present invention relates to a culture device for tissue cell suspension. The culture device includes a tissue cell culture body. The tissue cell culture body is a porous material. The porous material is formed by cavities classified into different levels according to the pore size of material and cavity walls surrounding to form the cavities. A lower level of small cavities is provided to surround the cavity wall which forms the upper level of large cavity. Cavities of each level are in communication with each other, and cavities between respective levels are also in communication with each other. The culture device further includes a swirler device provided therein. Such culture device can particularly facilitate normal and unrestricted growth of the suspension of cells in three-dimensional space, obstructing the formation of over-dense cell region or nutrient-rich region during the cell culturing.
Method of manufacturing a recombinant polypeptide
We have modified a commercially-available adherent cell culture bioreactor in several ways to increase productivity of cultured cells, while decreasing contamination risk. We found that modifying a commercially-available adherent cell culture bioreactor to provide for slower cell culture medium flow unexpectedly and dramatically increases the productivity of the cultured adherent cells. We also developed a new sampling manifold configuration and new way of taking samples, to reduce contamination risk.
UNIFORM CELL CULTURE SUBSTRATE FOR FIXED BED BIOREACTOR
A packed-bed bioreactor system for culturing cells is provided. The system includes a cell culture vessel having at least one interior reservoir, an inlet fluidly connected to the reservoir, and an outlet fluidly connected to the reservoir; and a cell culture matrix disposed in the reservoir and arranged as a fixed bed. The cell culture matrix includes a substrate configured for adhering cells thereto. The cell culture matrix maintains a uniform flow within the reservoir throughout the cell culture matrix.

A bioreactor system for culturing cells is provided. The system includes a cell culture vessel with at least one interior reservoir for flowing liquid media therethrough. The system also includes a cell culture matrix disposed in the reservoir, the cell culture matrix having a substrate with a polymer base and a polystyrene coating disposed on the polymer base.
SYSTEMS AND METHODS OF IMPEDANCE-BASED BIOMASS SENSING IN FIXED BED BIOREACTORS
A cell culture bioreactor is disclosed that has a cell culture vessel with an interior reservoir to contain a cell substrate, an inlet fluidly connected to the reservoir, and an outlet fluidly connect to the reservoir. The bioreactor also has a sensor system for measuring impedance within the reservoir. The sensor system includes a pair of electrodes that are spaced apart from each other with at least a portion of the cell substrate disposed between the pair of electrodes. Each electrode of the pair of electrodes includes a plurality of pores to allow at least one of cells and culture media to flow through the electrode.

This invention discloses a three-dimensional (3D) bioreactor for large scale expansion of immune cells and methods of use. The 3D bioreactor comprising at least one packed bed chamber comprising at least one porous scaffold; at least one porous scaffold coated with one or more extra cellular matrix protein (ECM); at least one container comprising a fluid media, the fluid media is configured to flow through said packed bed chamber with at least one porous coated scaffold; and at least one population of immune cells suspended in the fluid media, wherein, the at least one porous scaffold coated with said ECM is creates a stationary niche having low shear forces that imitate the natural growth environment of the immune cells and allows expansion of the immune cells population that flow through the coated porous scaffold in large scale.
