THREE-DIMENSIONAL BIOREACTORS WITH PARTICLE-FACILITATED ANTIBODY COATINGS
An apparatus and method for T-cell separation, activation, transduction and expansion. Three-dimensional (3D) bioreactors may be employed that include antibody coatings which are preferably antibody coated particles applied to the 3D bioreactor surface. Such 3D bioreactors can be employed for T-cell separation from peripheral blood mononuclear cells including attachment of T-cells to the 3D bioreactor surface for activation and transduction by lentivirus vectors to produce CAR T-cells.
Cell culture tool and method for producing cell culture tool
A cell culture tool configured to be produced in a simple facility to obtain a cell mass having a desired shape. The cell culture tool includes a cell culture base layer that contains a cell culture base. The cell culture base layer has a cell adhesive area to which a cell is adherable and a cell adhesion inhibitory area where cell adhesion is inhibited. The cell adhesion inhibitory area includes a modified product of the cell culture base.
System for the formation of fibrin foam
A device and method for forming fibrin foam, preferably utilizing a single container, comprising a base structured to be rotationally driven and including a separation chamber disposed and structured to receive a blood sample therein. The base includes a reaction chamber disposed in fluid communication with said separation chamber. The blood sample is separated into a plasma segment and a packed cell segment when subjected to sufficient centrifugation concurrent to the driven rotation of the base. According to structural features, and in some instances concurrent and continuous centrifugation, the plasma segment is directed from said separation chamber into said reaction chamber which includes sufficient quantities of gas and reactant composition to facilitate formation of fibrin foam therein concurrent to centrifugation. Further centrifugation of the fibrin foam and the inclusion of additional features such as a pressurized reaction chamber facilitate the formation of fibrin foam exhibiting varying chemical and/or physical properties.
BIOREACTOR AND RELATED METHODS
An apparatus for culturing cells includes a bioreactor. The bioreactor may be modular and may include in a chamber a fixed bed, such as an unstructured or structured fixed bed (such as a spiral bed) for culturing cells, with a return column arranged centrally within the chamber. The modular bioreactor may include a plurality of structured fixed bed arranged in a stacked configuration. The modular bioreactor may include an outer casing forming a space for conditioning (e.g., insulating, heating, cooling) at least a chamber in which cells are cultured. The bioreactor may also include an impeller with radially curved blades, and may also suspend the impeller so that it may move from side-to-side and align with an external drive. Related methods are also disclosed.
ROLLED ADHERENT CELL CULTURE SUBSTRATES FOR UNIFORM FLOW IN FIXED BED REACTORS
A cell culture matrix for culturing cells in a fixed bed reactor is provided. The cell culture matrix includes a first substrate material with an ordered and regular array of openings passing through the layer, the openings being separated by the substrate material having a physical structure that is substantially regular and uniform and that is configured for growing cell thereon. The physical structure and array of openings are configured such that, when the first substrate material is rolled into a rolled substrate matrix comprising rolled layers of the first substrate material, a variation in packing density of the rolled layers is less than about 20, less than about 10, less than about 5, or less than about 2 throughout the rolled substrate matrix.
FIXED BED SAMPLER AND RELATED METHODS
A sampler for use with a bioreactor for growing a cell culture is disclosed. In one embodiment, the bioreactor includes a structured fixed bed including a removable sample portion for recovering a sample of cells from the cell culture. Related apparatuses and methods are also disclosed.
