A medium composition for porcine pluripotent stem cells, containing bFGF, activin A, CHIR99021 and IWR-1 is disclosed. The composition is prepared from specific ingredients, excluding non-specific ingredients, thereby facilitating quality control, and thus is suitable for the study of porcine pluripotent stem cells. Also disclosed is a method for culturing, maintaining and preserving porcine pluripotent stem cells having the ability to form teratomas, by using the composition.

Method of banking stem cells
10959425 · 2021-03-30 · ·

A method of banking stem cells by harvesting stem cells from an individual and culturing the harvested stem cells to generate a P0 culture. The method includes storing a desired amount of P0 stem cells via cryopreservation and subculturing a portion of the P0 stem cells to generate a P1 culture. A desired amount of P1 stems cells can then be stored via cryopreservation and further generations can be stored as desired.

Drying formulation for hydrogel microcarriers

A method of making a cell culture article is provided. The method includes forming a microcarrier from a microcarrier composition comprising a polygalacturonic acid compound or an alginic acid compound, infiltrating the microcarrier with a drying formulation to form an infiltrated microcarrier, and drying the infiltrated microcarrier to form a dried microcarrier, wherein the drying formulation comprises at least one of a saccharide and a monovalent cation.

Culture medium formulations are described that are capable of large-scale expansion of stem cell-derived hepatocyte-like cells while sustaining the activity of those cells to maintain the phenotype and biological characteristics of normal hepatocytes.

METHOD FOR PRODUCING CARDIOMYOCYTES

The present invention chiefly aims to provide a process for directly inducing cardiomyocytes from somatic cells without performing artificial gene transfer, a cardiomyocyte obtained thereby, and a composition comprising a combination of chemical compounds capable of using for the said process. The present invention can include a process for producing a cardiomyocyte by inducing differentiation directly from a somatic cell, the process comprising a step of culturing the somatic cell in the presence of a MEK inhibitor and a cAMP inducer, and a cardiomyocyte obtained thereby, and then a composition for producing a cardiomyocyte by inducing differentiation directly from a somatic cell, the composition comprising a MEK inhibitor and a cAMP inducer. The cardiomyocytes obtained according to the present invention are useful in regenerative medicine and the like.

METHODS FOR PRODUCING HEPATOCYTES

Methods for producing hepatocytes from pluripotent human stem cells are disclosed herein. The stem cells are plated on a cell culture substrate comprising two laminins. The stem cells are then exposed to different cell culture mediums to induce differentiation. The resulting hepatocytes have higher metabolic capacity compared to hepatocytes cultured on different substrates.

STEM CELL PREPARATIONS AND APPLICATION IN THE PREPARATION OF DRUGS FOR THE TREATMENT OF OSTEOARTHRITIS

The present invention relates to the field of stem cell technology, and in particular to a stem cell preparation and its application in the preparation of drugs for the treatment of osteoarthritis (OA). The composition provided by the invention can significantly improve the activity of umbilical cord mesenchymal stem cells under cryopreservation conditions, and the stem cell preparation prepared therefrom has a good effect on repairing cartilage injury caused by OA, thereby playing a role in the treatment of OA.

METHODS AND COMPOSITIONS RELATING TO HEMATOPOIETIC STEM CELL EXPANSION, ENRICHMENT, AND MAINTENANCE

The methods and compositions described herein relate to producing, expanding, enriching, and/or maintaining hematopoietic stem cells ex vivo by treating the cells with an agent(s) that exhibits two or more activities selected from modulation of histone methylation; inhibition of TGF signaling; inhibition of p38 signaling; activation of canonical Wnt signaling; and modulation of histone acetylation. In some embodiments, the technology described herein relates to transplantation of hematopoietic stem cells.

A method for producing hepatocytes from hepatoblasts is provided. The method includes the step of culturing the hepatoblasts in a medium containing a compound selected from the group consisting of pregnenolone and an adrenergic agonist. The hepatoblasts can be obtained by culturing endodermal cells in a medium containing DMSO, and the endodermal cells can be obtained by culturing pluripotent stem cells in a medium containing Activin A and a GSK-3 inhibitor. Accordingly, a method for producing hepatocytes from pluripotent stem cells is also provided by employing the method of the present invention.

METHODS FOR INTEGRATION OF TRANSGENE DNA

Disclosed herein are methods of genome alteration, in particular genome editing in eukaryotic cells (e.g., mammalian cells), preferably, but not exclusively the integration of exogenous nucleic acids into the genome of a cell or a population of cells. Such methods include the modulation of cell cycle phases via external conditions such as physical separation, temperature, exposure to certain substances such as cell cycle modulators. Genome alteration is also effected via the use of enzymes such as nucleases and nickases and/or the modulation of DNA repair pathways.
