The invention also relates to probes and primers, and to antibodies against antigens from TiLV, and use of these reagents for detecting the presence or absence of TiLV in an animal. The invention also relates to iRNAs which target nucleic acid sequences of TiLV. The invention is also related to immunogenic compositions, including antibodies and vaccines, for inducing an immune response against TiLV in an animal. The invention is also related to gene constructs and cells comprising TiLV and isolated nucleic acids sequences and polypeptides thereof for use in developing prophylactic and therapeutic agents.
Prefusion PIV F immunogens and their use
Embodiments of a recombinant human Parainfluenza Virus (hPIV) F ectodomain trimer stabilized in a prefusion conformation are provided. Also disclosed are nucleic acids encoding the hPIV F ectodomain trimer and methods of producing the hPIV F ectodomain trimer. Methods for inducing an immune response in a subject are also disclosed. In some embodiments, the method can be a method for treating or inhibiting a hPIV infection in a subject by administering a effective amount of the recombinant hPIV F ectodomain trimer to the subject.
VACCINE CANDIDATES FOR HUMAN RESPIRATORY SYNCYTIAL VIRUS (RSV) HAVING ATTENUATED PHENOTYPES
Reported herein are presumptively de-attenuating mutations that are useful, either individually or in combinations that may include other known mutations, in producing recombinant strains of human respiratory syncytial virus (RSV) exhibiting attenuation phenotypes. Also described herein is a novel RSV construct, Min_L-NPM2-1(N88K)L, which exhibits an attenuated phenotype, is stable and is as immunogenic as wild type RSV. The recombinant RSV strains described here are suitable for use as live-attenuated RSV vaccines. Exemplary vaccine candidates are described. Also provided are polynucleotide sequences capable of encoding the described viruses, as well as methods for producing and using the viruses.

The present invention relates to compositions, immunogenic or vaccine compositions and pharmaceutical compositions for the prevention or treatment of canine atopic dermatitis (CAD). Furthermore, the invention provides methods for preventing or treating CAD. The present invention provides purification strategies for sterically demanding, i.e. large and pleomorphic, infectious virus particles or VLPs derived therefrom, preferably having a measles virus scaffold to yield fractions or compositions with a significantly reduced content of contaminating host cell DNA and a reduced content of further process-related impurities. Further provided are methods of propagating and purifying infectious virus particles having a measles virus scaffold suitable to provide a preparation having a strongly reduced content of contaminating host cell DNA and a reduced content of further process-related impurities for immunogenic or anti-tumor purposes. In addition, immunogenic and vaccine compositions based on the above methods are provided. Finally, there are provided immunogenic or vaccine compositions produced by the disclosed methods, which are suitable for use in immunogenic or prophylactic vaccination treatment of a subject in need thereof.
SELF-ASSSEMBLING NANOSTRUCTURE VACCINES
The present disclosure provides nanostructures and nanostructure-based vaccines. Some nanostructures of the present disclosure display antigens capable of eliciting immune responses to infectious agents such as bacteria, viruses, and pathogens. Some vaccines of the present disclosure are useful for preventing or decreasing the severity of infection with an infectious agent, including, for example and without limitation, lyme disease, pertussis, herpes virus, orthomyxovirus, paramyxovirus, pneumovirus, filovirus, flavivirus, reovirus, retrovirus, meningococcus, or malaria. The antigens may be attached to the core of the nanostructure either non-covalently or covalently, including as a fusion protein or by other means disclosed herein. Multimeric antigens may optionally be displayed along a symmetry axis of the nanostructure. Also provided are proteins and nucleic acid molecules encoding such proteins, vaccine compositions, and methods of administration.
LIQUID VACCINES OF LIVE ENVELOPED VIRUSES
The present invention describes a liquid vaccine composition of a live enveloped virus and a pharmaceutically acceptable carrier, whereby the carrier is a natural deep-eutectic solvent (NADES), and the vaccine has a water activity of less than about 0.8. The NADES provides a stabilisation of the sensitive virus for prolonged time and at ambient temperature. In general, the liquid vaccine compositions according to the invention, in different compositions for the various enveloped viruses, show remarkable capabilities of stabilisation. This overcomes the need for lyophilisation, a great economic benefit. Also the liquid nature of the vaccines facilitates administration to human or animal targets.
