PROCESS FOR WHOLE-CELL BIOSYNTHESIS OF STYRENE
20250146025 · 2025-05-08 ·

A process for producing styrene converts Z-phenylalanine using a first biocatalyst comprising a PAL enzyme (pheny-lalanine ammonia lyase from Rhodorotula glutinis EC 4.3.1.24) and a second biocatalyst comprising a Fdc1 enzyme (ferulic acid de-carboxylase from Aspergillus niger EC 4.1.1.102). The first and second biocatalysts are provided as whole-cell pellets or derivatives thereof. Styrene is produced by converting the L-phenylalanine to trans-cinnamic acid with the first biocatalyst and converting the trans-cinnamic acid to styrene with the second biocatalyst.

Production of geranyl diphosphate-derived compounds

Disclosed is yeast cells having peroxisomally localized GPP synthase and a peroxisomally localized enzyme that converts GPP into a monoterpenoids, cannabinoids, monoterpene indole alkaloids and prenylated aromatic compounds; or a precursor therefore, which yeast cells are capable of producing improved amounts of monoterpenoids, cannabinoids, monoterpene indole alkaloids and prenylated aromatic compounds, compared with the same yeast cells where the GPP synthase and the enzyme that converts GPP are located in the cytoplasm. Further disclosed is the use of the yeast cell for producing monoterpenoids, cannabinoids, monoterpene indole alkaloids and prenylated aromatic compounds.

Described herein are plant-derived haloacid dehalogenase-like (HAD-like) polypeptides having cyclic terpene synthase (TPS) activity, in particular suitable for use in biochemical methods of producing drimane-type sesquiterpenes, encompassing drimenol and/or albicanol and related compounds, like phosphorylated drimane-type sesquiterpene alcohols, in particular phosphorylated drimenol and/or albicanol compounds and derivatives. Also described herein are the coding nucleotide sequences of the TPS activity, corresponding expression constructs, recombinant hosts, and methods of preparing such polypeptides and mutants and variants thereof. Also described herein is a method of using such polypeptides, the method including using the polypeptides in the production of odorants, flavours and fragrance ingredients.
