High throughput cell free protein synthesis methods and systems are provided for expression of active, mature enzymes and proteins derived from zymogens and proproteins. Zymogens and proproteins are expressed in their mature, active form in a cell free expression system, without cleavage of a pro-sequence to produce the active polypeptide.

Variants of the transglutaminase enzyme of Streptomyces mobaraensis are provided. The disclosed variants exhibit at least about 2-fold increased enzymatic activity versus the wild-type enzyme. Methods are provided for increasing the self-life of products, such as personal care, household and industrial products, by incorporating an effective amount of the disclosed variant enzymes into the product. The transglutaminase variants may also be used to covalently bind functional ingredients, such as UV-blocking molecules, dyes, or pigments to proteins. The transglutaminase enzymes and functional ingredients may be incorporated into a cosmetic formulation for modifying skin, hair, or nail proteins or skin-derived proteins, such as collagen, keratin, and/or elastin.

The present invention relates to novel immobilized nanoflowers made of Transglutaminase (TGase), a method for their synthesis and applications thereof. The Transglutaminase nanoflowers of the present invention are catalytically more active without mass transfer limitation, stable and reusable, wherein the overall flower is in the nano-range, is homo-dispersed and small-sized, and wherein Transglutaminase functions both as a cross-linking enzyme and as a seeding template.
METHODS FOR PRODUCING STIRRED YOGURT
A method for producing stirred yogurt, whereby raw milk is treated with a protein glutaminase to form a modified milk prior to fermentation with a starter culture. After fermentation, a gel structure of the resulting yogurt is then disrupted to form the stirred yogurt. Under such a sequence, stirred yogurts are formed having sufficiently high viscosities without the need for thickeners or cross-linking enzymes.
Solid fibrinogen preparation
A fibrinogen preparation is provided which has an improved solubility, may be prepared within a short time and may be used rapidly in clinical set-up. A solid fibrinogen preparation comprising as a main ingredient fibrinogen and further containing the following components: albumin; a nonionic surfactant; a basic amino acid or a salt thereof; and at least two amino acids or a salt thereof selected from an acidic amino acid or a salt thereof and a neutral amino acid or a salt thereof. The solid fibrinogen composition of the present invention may be held on a medical material to form a supporting material holding fibrinogen. Besides, the supporting material holding fibrinogen may be combined with a component comprising as a main ingredient thrombin to provide a fibrin adhesive.
Treatment of cereal flour and semolina for consumption by celiac patients
The present invention relates to the use of an enzymatic method for treating flour, semolina and protein extracts derived from cereals known to stimulate a pathological immune response in patients affected by Celiac Disease (CD). Said treatment drastically reduces or completely eliminates the toxicity of gluten or similar products derived from other cereals. The method uses the catalytic activity of microbial transglutaminase and, in particular, its ability to bind proteins in cereal flour or semolina to an alkylated (C.sub.1-C.sub.4) derivative of lysine. After the treatment said proteins extracted from flour show a substantially lowered immuno-stimulatory activity in celiac patients T lymphocytes.
