Thiohydantoin derivatives and uses thereof

The present invention provides novel compounds (e.g., compounds of Formula (I)), and pharmaceutically acceptable salts, solvates, hydrate, polymorphs, co-crystals, tautomers, stereoisomers, isotopically labeled derivatives, prodrugs, and compositions thereof. Also provided are methods and kits comprising the inventive Compounds, or compositions thereof, for treating and/or preventing a fungal or protozoan infection, inhibiting the activity of a fungal or protozoan enzyme, killing a fungus or protozoan, or inhibiting the growth of a fungus or protozoan. The fungus may be a Candida species, Sacchawmyces species, or other pathogenic fungal species. The compounds of the invention may inhibit the activity of fungal or protozoan mitochondrial phosphate carrier protein.

Provided herein are compositions and systems comprising yeast-based biosensors (YBBs) and methods of use thereof. In particular, YBBs are provided for the detection and/or quantification of an analyte (e.g., peptide analyte) in a sample (e.g., a biological sample, environmental sample, etc.). In some embodiments, provided herein are diagnostic compositions, devices, and methods comprising yeast-based biosensors (YBBs) engineered to detect analytes (e.g., peptides (e.g., biomarker peptides, etc.), etc.). In some embodiments, YBBs comprise (a) recognition element (e.g., receptor (e.g., modified yeast receptor, etc.), etc.) on the exterior of the biosensor (e.g., for binding and or recognition of the analyte (e.g., peptide)), and (b) a reporter that signals binding or recognition of the analyte. In some embodiments, a recognition element is a cell surface receptor.

Time sensitive biopharmaceutical processing updates can be implemented using lateral flow devices to test on-site during manufacturing. Lateral flow testing for an analyte such as, for example, impurities (e.g., host cell proteins), during manufacturing of a biopharmaceutical drug product is completed within twenty-five minutes of collecting a sample from the manufacturing line. That is, all sample preparation, such as adding reagent or diluting the sample, contacting the prepared sample to a lateral flow device, and detecting presence or absence of the analyte is completed within twenty-five minutes (e.g., 20 minutes, 18 minutes, 15 minutes) from collection such that processing adjustments during manufacturing can be implemented.
