    Abstract

    The present invention relates to novel microspheres using an anionic polymer, a preparation method thereof, a composition, and the like, and the present invention is used for a method for performing a vascular embolization and/or necrosing cancer cells by blocking major blood vessels that supply nutrients to cancer cells to induce an embolic effect, and thus may be used for the treatment of patients with cancer.

    Claims

    1. A composition for the preparation of microspheres consisting of an anionic polymer and a multivalent metal compound salt as active ingredients.

    2. The composition of claim 1, wherein the anionic polymer is selected from the group consisting of carboxymethyl cellulose (CMC) or a salt thereof; dextran sulfate (DS) or a salt thereof; and a mixture thereof

    3. The composition of claim 1, wherein the microspheres comprise carboxymethyl cellulose and dextran sulfate.

    4. The composition of claim 3, wherein the carboxymethyl cellulose or a salt thereof and the dextran sulfate or a salt thereof are comprised at a mass ratio of 0.5 to 20:1.

    5. The composition of claim 1, wherein the multivalent metal compound salt is one or more divalent to tetravalent metal compound salt selected from the group consisting of Zr.sup.2+, Zr.sup.4+, Fe.sup.2+, Fe.sup.3+, Ca.sup.2+, Al.sup.3+, Cu.sup.2+, and Zn.sup.2+.

    6. The composition of claim 1, the microspheres are for use in performing a vascular embolization.

    7. The composition of claim 1, wherein the cationic drug is loaded into the microspheres.

    8. The composition of claim 7, wherein the cationic drug is one or more selected from the group consisting of doxorubicin, procainamide, digoxin, quinidine, trimethoprim, cimetidine, vancomycin, irinotecan, daunorubicin, epirubicin, diphenhydramine, memantine, oxycodone, pyrilamine, tramadol and pharmaceutically acceptable salt thereof

    9. The composition of claim 7, wherein the cationic drug and the microsphere are comprised at a mass ratio of 1:1 to 10.

    10. The composition of claim 1, wherein the microspheres are for use in treating cancer.

    11. The composition of claim 1, wherein the microsphere have a diameter of 10 to 700 m.

    12. A method for preparing microsphere consisting of an anionic polymer and a multivalent metal compound salt, the method comprising the following steps: forming an emulsion by mixing a solution obtained by adding a water phase in which a polymer is dissolved to an oil phase with a solution in which a non-ionic surfactant is dissolved in a water phase; forming microsphere by adding an aqueous multivalent metal compound salt solution to the emulsion; and purifying target size of microparticle by selective pore of sieve.

    13. The method of claim 11, wherein the anionic polymer is selected from the group consisting of carboxymethyl cellulose or a salt thereof; dextran sulfate or a salt thereof; and a mixture thereof.

    14. The method of claim 11, wherein the oil phase is selected from the group consisting of mineral oil, vegetable oils, heavy chain triglycerides and a mixture thereof.

    15. The method of claim 11, wherein the non-ionic surfactant is selected from the group consisting of polyvinyl alcohol, polyalkylene glycol, polyvinyl pyrrolidone, an alkyl polyglycoside, t-octylphenoxy polyethoxy ethanol, a poloxamer, polysorbate, poly(N-vinylacetamide), polyvinyl butyral, and a mixture thereof.

    16. A method for preparing the composition for a vascular embolism and treating cancer of claim 1, the method comprising the following steps: forming an emulsion by mixing a solution obtained by adding a water phase in which a polymer is dissolved to an oil phase with a solution in which a non-ionic surfactant is dissolved in a water phase; forming microspheres by adding an aqueous multivalent metal compound salt solution to the emulsion; purifying target size of microparticle by selective pore of sieve; and mixing one or more cationic drug with the microsphere.

    Example 2. Preparation of Microparticles Using W/O Emulsion

    [0210] A CMC solution, an iron chloride (FeCl.sub.3) solution, and a dextran sulfate (DS) powder were each prepared according to the composition ratios in the following Table 2.

    Example 4. Preparation of Microspheres Through Additional Nanoprocessing After Preparation of Microparticles Using W/O Emulsion

    [0216] The preparation of microparticles was implemented by the following method. To prepare a water phase solution, 2 g of PVA and 1 g of CMC were each dissolved in 100 mL of distilled water. Then, 0.5 g of DS was measured, added to the CMC solution, and then dissolved. To prepare a cross-linking agent solution, 0.5 g of iron (III) chloride (FeCl.sub.3) was dissolved in 100 mL of distilled water.

    [0217] To prepare a W/O emulsion, a 2 L beaker was filled with 800 mL of the MCT oil, and all of the water phase solutions were added while being stirred at room temperature at 17,000 rpm using a homogenizer. After the mixture was stirred under the same conditions for 10 minutes, the cross-linking agent solution was added dropwise thereto, and the resulting mixture was further stirred for about 30 minutes.

    [0218] Microparticles were separated using the following experimental method: The reaction solution was transferred to a separatory funnel and allowed to stand for about 2 hours. The lower layer liquid (water phase) in which the yellow solid was present in the separatory funnel was collected and yellow particle pellets were produced using a centrifuge. The supernatant was discarded, and a white material was dissolved and removed using ethanol. A ratio of doxorubicin hydrochloride: particles=25 mg: 6 mg was adjusted by putting a completely dissolved doxorubicin hydrochloride sucrose solution into a 1.25-mL 5%-sucrose solution in which 60 mg of microspheres were dispersed, and the resulting mixture was additionally mixed for about 30 minutes after an initial bath type sonication treatment for 1 minute.

    [0223] 500 L of the microsphere suspension loaded with the doxorubicin hydrochloride was put into a CelluSep membrane (MW: 15,000, Width: 32 mm, Wall thickness: 24 m), and both sides were blocked with clips. 50 mL of a buffer (PVA 0.4%, Tween 0.1% in lactic acid pH=2.7, and 37 C.) was put into a vial and stirred at 200 rpm, and the temperature was increased to 37 C. The membrane sample was put thereinto and an elution test was performed.     Example 6. In Vitro Cytotoxicity Evaluation

    [0225] Viability analysis was performed on Huh7 liver cancer cells to evaluate the toxicity of the prepared microspheres. Three groups were evaluated: doxorubicin hydrochloride (dox), particles loaded with doxorubicin (032d), and particles not loaded with doxorubicin (032).

    Example 7. Preparation of Mixed Emulsion with Lipiodol and Confirmation of Passage Through Microcatheter

Then, 10 mL of a Lipiodol ultra solution (Guerbet Korea) was prepared in a separate syringe. An emulsion was prepared by placing the two solutions in a 3-way cock and alternately pressing the syringes. An image of the formed emulsion is illustrated in FIG. 8.

CMC and DS solution was stirred for 2 h at room temperature. And 2.5 g of Zirconyl chloride octahydrate(ZrOCl.sub.2.Math.8H.sub.2O) as a cross linker was fully dissolved to 100 g of DI water at room temperature. And CMC and DS mixture was slowly dropwise into the ZrOCl.sub.2.Math.8H.sub.2O solution. CMC and DS microsphere were cross-linked for 2 hours and washed by DI water. Appearance of CMC and DS microparticles was analyzed by microscopy (FIG. 10). Transparent microspheres were observed. To bind doxorubicin hydrochloride to microsphere, doxorubicin hydrochloride solution (Drug product) at the concentration of 2 mg/mL was mixed to microsphere (N=13) and stored at room temperature for overnight. And appearance of doxorubicin bound microsphere was observed by microscopy. And binding doxorubicin was confirmed by the change of color to red (FIG. 11).

    Example 9. Dissociation of Microsphere and Doxorubicin Bound Microsphere

    [0235] Empty microsphere and doxorubicin bound microsphere were dissociated in phosphate buffered saline (PBS, pH 7.4). Each microsphere was separately stored in vial containing 100 mL of DW. Vial was stored at the room temperature. And dissociation was observed for 7 days (FIGS. 12, 13).

    Example 10. Synthesis of CMC-DS Microsphere Dependent on Cross-Linkers

    [0236] CMC-DS microspheres were synthesized with various cross-linkers. The method to fabricate microsphere was followed to the method in Example 8, except for cross-linker solution. And all of cross-linker experiments were performed independently.

CMC-DS mixtures were dropwised to each cross-linkers and stirred for 2 hours. After being cross-linked, CMC-DS microsphere was washed by DI water. Appearance of each CMC-DS microsphere were observed by microscope (FIG. 14).

    Example 12. Control of Size Distribution

    [0240] CMC-DS microsphere was fabricated with the control of size distribution. Fabrication of CMC-DS microsphere for the control of size distribution was achieved by method of emulsification and filtration. Firstly, 2% of CMC and 0.5% of DS was dissolved to the water. And CMC and DS mixture was dropwised to MCT oil containing 4% of span 80. Aqueous and oil phase ratio was 25% and homogenized with the 3000 rpm for 10 min. this CMC-DS emulsion(W/O) was additionally dropwised at 3 mL/min to the 2.5% of ZrOCl.sub.2.Math.8H.sub.2O solution as cross-linker.
