    Claims

    1. A compound of formula (II) or a pharmaceutically acceptable salt thereof, ##STR00067## wherein R.sub.1 is selected from R.sub.2, ##STR00068## and -L-R.sub.3; R.sub.2 is selected from cyclopentyl, C.sub.4-6 bicycloalkyl, and 4- to 5-membered heterocycloalkyl, and the cyclopentyl, C.sub.4-6 bicycloalkyl, and 4- to 5-membered heterocycloalkyl are each independently and optionally substituted by 1, 2, or 3 R.sub.a; R.sub.3 is selected from C.sub.3-6 cycloalkyl and 4- to 5-membered heterocycloalkyl, and the C.sub.3-6 cycloalkyl and 4- to 5-membered heterocycloalkyl are each independently and optionally substituted by 1, 2, or 3 R.sub.b; L is selected from O, OCH.sub.2, CH(R.sub.c), and C(R.sub.dR.sub.e); R.sub.a and R.sub.b are each independently selected from halogen, CN, C.sub.1-3 alkyl, and C.sub.1-3 alkoxy, and the C.sub.1-3 alkyl and C.sub.1-3 alkoxy are each independently and optionally substituted by 1, 2, or 3 R.sub.f; R.sub.c, R.sub.d, and R.sub.e are each independently selected from halogen and C.sub.1-3 alkyl; R.sub.f is selected from halogen; R.sub.4 is selected from halogen and C.sub.1-3 alkyl; T.sub.1 is selected from CH and N; hetero in the heterocycloalkyl represents 1, 2, or 3 heteroatoms or heteroatom groups, each independently selected from O, NH, S, and N.

    2. The compound or the pharmaceutically acceptable salt thereof according to claim 1, wherein R.sub.a and R.sub.b are each independently selected from F and methoxy.

    3. The compound or the pharmaceutically acceptable salt thereof according to claim 1, wherein R.sub.c, R.sub.d, and R.sub.e are each independently selected from F and methyl.

    4. The compound or the pharmaceutically acceptable salt thereof according to claim 1, wherein R.sub.f is selected from F.

    5. The compound or the pharmaceutically acceptable salt thereof according to claim 1, wherein R.sub.2 is selected from ##STR00069## and the ##STR00070## are each independently and optionally substituted by 1, 2, or 3 R.sub.a.

    6. The compound or the pharmaceutically acceptable salt thereof according to claim 5, wherein R.sub.2 is selected from ##STR00071##

    7. The compound or the pharmaceutically acceptable salt thereof according to claim 1, wherein R.sub.3 is selected from ##STR00072## and the ##STR00073## are optionally substituted by 1, 2, or 3 R.sub.b.

    9. The compound or the pharmaceutically acceptable salt thereof according to claim 1, wherein L is selected from O, OCH.sub.2, and C(CH.sub.3).sub.2.

    17. The method according to claim 16, wherein the disease related to soluble guanylate cyclase activators is chronic kidney disease.

    DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENT

    [0094] The present disclosure is described in detail by the examples below, but it does not mean that there are any adverse restrictions on the present disclosure. The present disclosure has been described in detail herein, and its specific examples have also been disclosed; for one skilled in the art, it is obvious to make various modifications and improvements to the examples of the present disclosure without departing from the spirit and scope of the present disclosure.

    Step 1: Synthesis of Compound WX001_2

    [0097] tert-Butyl diethylphosphonoacetate (5.14 g, 20.38 mmol) was dissolved in tetrahydrofuran (30 mL) at room temperature under nitrogen atmosphere. The reaction system was cooled to 0? C., and potassium tert-butoxide (2.52 g, 22.48 mmol) was added thereto. After the reaction mixture was stirred for 30 minutes, WX001_1 (2 g, 20.38 mmol) was slowly added dropwise thereto. Once the addition was complete, the reaction system returned to room temperature and was stirred for 12 hours. After the reaction was complete, the reaction mixture was diluted with water (50 mL), and then extracted with ethyl acetate (60 mL?3). The organic phases were combined. The organic phase was washed with saturated brine (60 mL?2), dried over anhydrous sodium sulfate, filtered, and then concentrated under reduced pressure to remove the solvent. The resulting crude product was separated by column chromatography (eluent: petroleum ether/ethyl acetate=1/0 to 1/1, v/v) to obtain compound WX001_2. .sup.1H NMR (400 MHz, DMSO_d.sub.6) 6: 6.75 (dd, J=8.0, 15.5 Hz, 1H), 5.75-5.65 (m, 1H), 2.62-2.52 (m, 1H), 1.83-1.72 (m, 2H), 1.65-1.53 (m, 4H), 1.42 (s, 9H), 1.36-1.28 (m, 2H).

    Step 2: Synthesis of Compound WX001_4

    [0098] 2,2-Bis(diphenylphosphino)-1,1-binaphthalene (18.24 mg, 29.29 ?mol) and chloro(1,5-cyclooctadiene)rhodium(I) dimer (62.80 mg, 127.37 ?mol) were dissolved in tetrahydrofuran (10 mL) at room temperature under nitrogen atmosphere. The reaction system was stirred at room temperature for 15 minutes. Compound WX001_3 (1.42 g, 5.60 mmol) was dissolved in isopropanol (5 mL) and tetrahydrofuran (10 mL). Compound WX001_2 (1.00 g, 5.09 mmol), potassium hydroxide (343.00 mg, 6.11 mmol), and 1,5-cyclooctadiene (55.11 mg, 509.46 ?mol) were sequentially added thereto. The reaction system was heated to 60? C., added with the prepared catalyst, and stirred at 60? C. for 9 hours. After the reaction was complete, the reaction mixture was cooled to room temperature, diluted with water (150 mL), and then extracted with ethyl acetate (150 mL?3). The organic phases were combined. The organic phase was washed with saturated brine (100 mL?2), dried over anhydrous sodium sulfate, filtered, and then concentrated under reduced pressure to remove the solvent. The resulting crude product was separated by column chromatography (eluent: petroleum ether/ethyl acetate=1/0 to 1/1, v/v) to obtain compound WX001_4. .sup.1H NMR (400 MHz, CDCl.sub.3) ?: 7.12 (d, J=8.1 Hz, 1H), 6.60 (d, J=2.0 Hz, 1H), 6.52 (dd, J=2.0, 8.3 Hz, 1H), 3.98 (s, 2H), 2.73-2.61 (m, 2H), 2.45-2.35 (m, 1H), 1.97-1.78 (m, 2H), 1.70-1.61 (m, 1H), 1.59-1.49 (m, 2H), 1.45-1.35 (m, 2H), 1.28-1.25 (m, 9H), 1.08-0.96 (m, 1H), 0.91-0.82 (m, 1H).

    Step 3: Synthesis of Compound WX001_7

    [0099] Diisopropylamine (31.52 g, 311.45 mmol, 44.02 mL) was placed in a dry reaction flask at room temperature under nitrogen atmosphere. Tetrahydrofuran (350 mL) was added to dissolve the compound. The reaction system was cooled to ?70? C., and a solution of n-butyllithium in hexane (130.00 mL, 324.99 mmol, 2.5 M) was added thereto. The reaction system was stirred at ?70? C. for 30 minutes, then warmed to ?40? C., and stirred for another 30 minutes. The reaction system was cooled to ?70? C., and WX001_6 (50 g, 270.83 mmol) dissolved in tetrahydrofuran (120 mL) was added thereto. The reaction system was stirred at ?70? C. for 1 hour. Trifluoroacetone (45.52 g, 406.24 mmol) dissolved in tetrahydrofuran (150 mL) was then added, and the reaction system was stirred at ?70? C. for another 2 hours. After the reaction was complete, the reaction system was warmed to 0? C., added with 120 mL of 5 N dilute hydrochloric acid to quench the reaction, and then stirred at room temperature for 30 minutes. The reaction mixture was concentrated under reduced pressure to remove most of the organic solvent, and the residue was diluted with ethyl acetate (300 mL). The phases were separated, and the organic phase was collected. The aqueous phase was extracted with ethyl acetate (200 mL?3), and the organic phases were combined. The organic phase was washed with saturated brine (200 mL?2), dried over anhydrous sodium sulfate, filtered, and concentrated under reduced pressure to remove the solvent to obtain compound WX001_7.

    Step 4: Synthesis of Compound WX001_8

    [0100] WX001_7 (80.35 g, 270.84 mmol) was dissolved in pyridine (160 mL) at room temperature under nitrogen atmosphere. To the reaction system at room temperature was added phosphorus oxychloride (49.83 g, 325.01 mmol). The reaction system was stirred at 110? C. for 12 hours. After the reaction was complete, the reaction system was cooled to room temperature, poured into 200 mL of water to quench the reaction, added with 6 N dilute hydrochloric acid to adjust the pH to around 3, and diluted with dichloromethane (400 mL). The phases were separated, and the organic phase was collected. The aqueous phase was extracted with dichloromethane (300 mL?3), and the organic phases were combined. The organic phase was washed with saturated brine (200 mL?2), dried over anhydrous sodium sulfate, filtered, and then concentrated under reduced pressure to remove the solvent. The resulting crude product was separated by column chromatography (eluent: petroleum ether/ethyl acetate=1/0 to 9/1, v/v) to obtain a mixture of compounds WX001_8 and WX001_9. The mixture was then separated by column chromatography (eluent: petroleum ether/ethyl acetate=1/0 to 9/1, v/v) to obtain compound WX001_8.

    Step 5: Synthesis of Compound WX001_9

    [0101] Diisopropylamine (7.99 g, 78.95 mmol) was placed in a dry reaction flask and dissolved in tetrahydrofuran (50 mL) at room temperature under nitrogen atmosphere. The reaction system was replaced with nitrogen three times and then cooled to ?60? C. A solution of n-butyllithium in hexane (71.77 mmol, 28.71 mL, 2.5 M) was slowly added thereto. The reaction system was warmed to ?40? C. and stirred at ?40? C. for 30 minutes. Afterwards, the reaction system was cooled to ?65? C., and WX001_8 (10 g, 35.89 mmol) dissolved in tetrahydrofuran (50 mL) was added dropwise thereto. The reaction system was stirred at ?65? C. for another 1.5 hours. After the reaction was complete, a solution of acetic acid (8.62 g, 143.55 mmol) in tetrahydrofuran (40 mL) was added to the reaction system to quench the reaction. The reaction mixture was then stirred at room temperature for 2 hours. The reaction mixture was diluted with water (80 mL) and ethyl acetate (80 mL), and then the phases were separated. The organic phase was collected and the aqueous phase was extracted with ethyl acetate (80 mL?3). The organic phases were then combined. The organic phase was washed with saturated brine (80 mL?2), dried over anhydrous sodium sulfate, filtered, and then concentrated under reduced pressure to remove the solvent. The resulting crude product was separated by column chromatography (eluent: petroleum ether/ethyl acetate=1/0 to 9/1, v/v) to obtain compound WX001_9. The crude product was directly used in the next step.

    Step 6: Synthesis of Compound WX001_10

    [0102] Pt/C (0.57 g, purity: 5%) was added to a dry hydrogenation flask and wetted with methanol (60 mL) at room temperature under argon atmosphere. WX001_9 (5.7 g, 20.46 mmol) was then added to the reaction system. The reaction system was stirred at 50? C. under hydrogen atmosphere of 50 psi for 12 hours. Upon completion of the reaction, the reaction system was filtered through diatomite. The filter cake was washed with methanol (60 mL?5), and the filtrates were combined and concentrated under reduced pressure to obtain the residue. Compound WX001_10 was obtained.

    Step 7: Synthesis of Compound WX001_11

    [0103] WX001_10 (5.8 g, 20.66 mmol) was dissolved in methanol (60 mL) at room temperature under nitrogen atmosphere. To the reaction system was then added sodium methoxide (3.35 g, 61.99 mmol). The reaction system was stirred at 80? C. for 12 hours. Additional sodium methoxide (1.12 g, 20.66 mmol) was added thereto, and the reaction system was stirred at 80? C. for another 12 hours. The reaction mixture was heated to 90? C. and stirred for 12 hours. After the reaction was complete, the reaction mixture was cooled to room temperature, concentrated under reduced pressure to remove most of the organic solvent, diluted with water (60 mL), and then extracted with methyl tert-butyl ether (70 mL). The organic phase was discarded, the aqueous phase was added with 3 N dilute hydrochloric acid to adjust the pH to 2 to 3 and extracted with ethyl acetate (80 mL?3), and the organic phases were combined. The organic phase was washed with saturated brine (50 mL?2), dried over anhydrous sodium sulfate, filtered, and then concentrated under reduced pressure to remove the solvent. Compound WX001_11 was obtained.

    Step 8: Synthesis of Compound WX001_5

    [0104] Quinine (3.51 g, 10.83 mmol) was dissolved in ethanol (36 mL) at room temperature under nitrogen atmosphere. The reaction system was heated to 80? C., and WX001_11 (3.85 g, 14.44 mmol), dissolved in ethanol (19 mL), was gradually added dropwise thereto. The reaction system was stirred at 80? C. for 10 minutes. A white solid precipitated. The reaction system was slowly cooled to 0? C. and stirred at 0? C. for 1 hour. The reaction mixture was filtered, and the filter cake was washed with 5 mL of water. The solid was collected. The collected solid was dissolved in methanol (157 mL) and water (20 mL). The reaction system was then heated to 100? C. and stirred for 30 minutes. It was subsequently slowly cooled to room temperature, resulting in the precipitation of a white solid. The reaction mixture was filtered, and the filter cake was washed with 10 mL of water. The resulting filter cake was collected. The filter cake was dissolved in ethanol (120 mL) and water (20 mL). The reaction system was then heated to 100? C. and stirred for 30 minutes. It was subsequently slowly cooled to room temperature, resulting in the precipitation of a white solid. The filter cake was washed with 10 mL of water, and the solid was collected. The solid was suspended in water (25 mL) and hydrochloric acid (6 M, 3.25 mL) was added thereto. The reaction system was stirred at room temperature for 2.5 hours. The white solid was then filtered and the filter cake was washed with 1 mL of water. The solid was collected, and the filter cake was concentrated under reduced pressure to remove the solvent. Thus compound WX001_5 was obtained.

    Step 9: Synthesis of Compound WX001_12

    [0106] Compound WX0015 (150 mg, 562.55 ?mol) was dissolved in dichloromethane (3 mL) at room temperature under nitrogen atmosphere. N,N-dimethylformamide (411.19 ?g, 5.63 ?mol) was added thereto, and the reaction system was cooled to 0? C. Oxalyl chloride (107.11 mg, 843.82 ?mol) was then added dropwise thereto. The reaction system was stirred at 25? C. for 1 hour, and concentrated under reduced pressure to remove the solvent. The residue was dissolved in dichloromethane (3 mL), and a solution of compound WX001_4 (218.62 mg, 675.06 ?mol) in dichloromethane (2 mL) was added dropwise thereto, along with N,N-diisopropylethylamine (145.41 mg, 1.13 mmol). The reaction system was stirred at room temperature for 2 hours. After the reaction was complete, the reaction mixture was concentrated under reduced pressure to remove the solvent.     Step 10: Synthesis of Compound WX001

    [0107] Compound WX001_12 (150 mg, 262.02 ?mol) was dissolved in ethyl acetate (3 mL) in a pre-dried reaction flask at room temperature under nitrogen atmosphere. Hydrochloric acid in ethyl acetate (4 M, 3 mL) was added thereto, and the reaction system was stirred at room temperature for 10 hours. After the reaction was complete, the reaction mixture was concentrated under reduced pressure to remove the solvent. The resulting residue was separated by preparative HPLC (mobile phase: acetonitrile/water; acidic system: 0.04% HCl) to obtain compound WX001.     Step 1: Synthesis of Compound WX004_2

    [0115] Compound WX004_1 (7.8 g, 27.52 mmol) was dissolved in methanol (80 mL) at room temperature under nitrogen atmosphere. Iron powder (7.68 g, 137.59 mmol) was added thereto, followed by a dropwise addition of a solution of ammonium chloride (11.78 g, 220.14 mmol) in water (40 mL). The reaction system was stirred at 70? C. for 12 hours. After the reaction was complete, the reaction mixture was filtered. The filter cake was washed with methanol (50 mL?2). The filtrate was collected, and concentrated under reduced pressure to remove the solvent. The reaction mixture was diluted with water (50 mL) and then extracted with ethyl acetate (100 mL?2). The organic phases were combined. The organic phase was washed with saturated brine (50 mL?2), dried over anhydrous sodium sulfate, filtered, and then concentrated under reduced pressure to remove the solvent. The resulting residue was purified by column chromatography (eluent: petroleum ether/ethyl acetate=1/0 to 5/1, v/v) to obtain compound WX004_2.

    Step 2: Synthesis of Compound WX004_3

    [0116] To N,N-dimethylformamide (50 mL) was added sodium hydride (1.10 g, 27.42 mmol, purity: 60%) in batches at 0? C. under nitrogen atmosphere. A solution of compound WX004_2 (2.78 g, 10.97 mmol) in N,N-dimethylformamide (30 mL) was then slowly added dropwise to the reaction system. After the reaction system was stirred at 0? C. for 0.5 hours, 4-methoxybenzylchloride (4.09 g, 26.10 mmol, 3.55 mL) was added dropwise thereto. Following the addition, the reaction system was warmed to 25? C. and stirred for 12 hours. After the reaction was complete, the reaction system was slowly pouring into water (50 mL) to quench and diluted with ethyl acetate (100 mL), and the phases were separated. The organic phase was collected, and the aqueous phase was extracted with ethyl acetate (50 mL?3). The organic phases were then combined. The organic phase was washed with saturated brine (50 mL?2), dried over anhydrous sodium sulfate, and filtered. The filtrate was concentrated under reduced pressure to remove the solvent. The resulting residue was purified by column chromatography (eluent: petroleum ether/ethyl acetate=1/0 to 5/1, v/v) to obtain compound WX004_3. .sup.1H NMR (400 MHz, CDCl.sub.3) ?: 7.36-7.23 (m, 6H), 7.18 (d, J=8.4 Hz, 1H), 6.93 (d, J=8.5 Hz, 4H), 4.18 (s, 4H), 3.89 (s, 6H).

    Step 3: Synthesis of Compound WX004_5

    Test Example 2: In Vitro Hepatocyte Metabolic Stability Study

    1. Experimental Objective: The Aim of the Study was to Assess the Stability of the Compounds in Hepatocytes of Different Species.

    2. Experimental Steps:

    [0225] A variety of 96-well precipitation plates were prepared and labeled as T0, T15, T30, T60, T90, T0-MC, T90-MC, and blank matrix, respectively. Revival medium and incubation medium were taken in advance and pre-warmed in a 37? C. water bath. Hepatocytes of different species, stored in a liquid nitrogen tank, were taken and immediately immersed in a 37? C. water bath for approximately 90 seconds. Once partially thawed, cells were transferred to centrifuge tubes containing 40 mL of revival medium, and gently inverted to resuspend the cells in the revival medium. At room temperature, cells were centrifuged at 100?g for 5 minutes. The supernatant was discarded, and the hepatocytes were resuspended in an appropriate volume of incubation medium. Viability was calculated using trypan blue staining. 198 ?L of hepatocyte suspension (0.51?10.sup.6 cells/mL) was added to pre-warmed incubation plates. For the control group, 198 ?L of hepatocyte-free incubation medium was added to T0-MC and T120-MC plates. All plates were pre-incubated for 10 minutes in a 37? C. incubator. 2 ?L of test and control compound working solutions were then added, mixed thoroughly, and the incubation plates were immediately placed in a shaker within the incubator. The timer was initiated to start the reaction. Two duplicate samples were prepared at each time point for each compound. Incubation conditions were set at 37? C., saturated humidity, and 5% CO.sub.2. In the test system, the test compound had a final concentration of 1 ?M, the control compound had a final concentration of 3 ?M, the hepatocyte had a final concentration of 0.5?10.sup.6 cells/mL, and the total organic solvent had a final concentration of 0.96%, of which DMSO had a final concentration of 0.1%. At the end of the incubation at the corresponding time point, the incubation plate was removed, and 25 ?L of the compound and control compound-cell mixture was transferred to a sample plate containing 125 ?L of stop solution (acetonitrile solution containing 200 ng/mL of tolbutamide and Labetalol). For the blank sample plate, 25 ?L of hepatocyte-free incubation medium was directly added. All sample plates were sealed and shaken on the shaker at 600 rpm for 10 minutes, and then centrifuged at 3220?g for 20 minutes. The supernatant of test and control samples was diluted with ultrapure water at a ratio of 1:3. All samples were mixed well and analyzed using LC/MS/MS methods.
