Aerobic fermentation systems and methods
A system for aerobic fermentation includes a vessel, an aeration system including a gas sparger fluidly coupled to the vessel to introduce a compressed gas to an internal volume of the vessel, and a recirculation loop fluidly coupled to an outlet of the vessel. The recirculation loop includes an eductor fluidly coupled to an oxygen-containing gas source, a static mixer downstream of the eductor, a heat exchanger downstream of the eductor, and a distributor downstream of the heat exchanger. The distributor is fluidly coupled to the vessel. The aeration system provides mixing and oxygen mass transfer to the fermentation composition in the vessel. The fermentation composition passes through the eductor, static mixer, heat exchanger, and distributor of the recirculation loop, and back into the vessel. Oxygen is transferred from an oxygen containing gas to the fermentation composition and heat is removed from the fermentation composition in the recirculation loop.
Aseptic cell processing and production with no chemical biocides
A method and apparatus of aseptic processing and production of cells in a non-sterile enclosure apparatus without chemical biocides is disclosed, by controlling the level of humidity throughout the enclosure to 25% relative humidity (RH) or less, and preferably 20% or 15% or less RH. In addition, the temperature is controlled to 37° C., and consistent gas flow is maintained the enclosure. Colony forming units from microbial contamination detected by environmental monitoring within the enclosure are significantly reduced in this method.

Automated cell culture incubator
Aspects relate to an automated cell culture incubator and to methods for using such an incubator. In one aspect, the cell culture incubator includes an incubator cabinet having a transfer chamber and an internal chamber that are arranged to form an airlock configuration. In some embodiments, objects placed into the incubator cabinet are sterilized using ozone produced by an ozone generator. The incubator may include one or more transfer devices that move objects between the transfer chamber and the internal chamber.

In some embodiments, an automated closed apparatus for cell therapy manufacturing includes: a master device having a master controller for processing control programs for a variety of cell types; an input device fluidly coupled to the master device, wherein the input device is configured to feed an initial plurality of cells to the master device; one or more auxiliary devices each configured to perform one or more cell therapy manufacturing steps to the initial plurality of cells to form a final plurality of cells; and an output device coupled to the master device configured to collect the final plurality of cells from the master device.
