Functional sex-reversal of decapod crustacean females
The present invention provides a primary cell culture which combines a cell culture medium and cells derived from a hypertrophied androgenic gland (AG) of a decapod crustacean. The invention also provides methods for obtaining an all-female progeny by initially injecting/transplanting the primary cell culture to a genetic-female to obtain a male-Neo-male.
Blastoid, cell line based artificial blastocyst
A method for making an at least double layered cell aggregate and/or an artificial blastocyst, and/or a further-developed blastoid termed blastoid, by forming a double layered cell aggregate from at least one trophoblast cell and at least one pluripotent and/or totipotent cell, and culturing the aggregate to obtain an artificial blastocyst having a trophectoderm-like tissue that surrounds a blastocoel and an inner cell mass-like tissue. The cell aggregate can be formed from toti- or pluripotent stem cell types, or induced pluripotent stem cell types, in combination with trophoblast stem cells. Formation of a blastoid can be achieved by culturing the cell aggregate in a medium preferably comprising one or more of a Rho/ROCK inhibitor, a Wnt pathway modulator, a PKA pathway modulator, a PKC pathway modulator, a MAPK pathway modulator, a STAT pathway modulator, an Akt pathway modulator, a Tgf pathway modulator and a Hippo pathway modulator. Also, a method for growing an at least double layered cell aggregate into an artificial blastocyst, and into a further-developed blastoid, a foetus or a live animal and an in vitro cell culture comprising the mentioned compounds and/or cell aggregates.
Bos taurus variety ‘JE840003146074527’ and methods of use therof
The disclosure relates to Bovine germplasm of Bos taurus variety JE840003146074527. Included in the present disclosure are cells comprising the Bovine variety JE840003146074527. Also provided by the present disclosure are tissue cultures of cells, animals obtained from said cells, and parts thereof, including F1 spermatozoa. The disclosure further provides for methods of breeding, selecting, and using the germplasm to improve existing commercial cattle herds generated from in vitro fertilization methods and progeny cattle obtained from in vitro fertilization and implantation and artificial insemination methods.
METHODS AND COMPOSITIONS FOR TARGETED GENETIC MODIFICATIONS AND METHODS OF USE
Methods and compositions are provided for generating targeted genetic modifications on the Y chromosome or a challenging target locus. Compositions include an in vitro culture comprising an XY pluripotent and/or totipotent animal cell (i.e., XY ES cells or XY iPS cells) having a modification that decreases the level and/or activity of an Sry protein; and, culturing these cells in a medium that promotes development of XY F0 fertile females. Such compositions find use in various methods for making a fertile female XY non-human mammal in an F0 generation.
HYDROGEL INCLUSION COMPLEX INCLUDING PHYSIOLOGICALLY ACTIVE MATERIAL BOUND TO THERMOSENSITIVE POLY(PHOSPHAZENE) BY HOST-GUEST INTERACTION USING BETA-CYCLODEXTRIN AND USE THEREOF
Provided is a hydrogel composition including thermosensitive poly(phosphazene) to which a plurality of hydrophobic amino acids, hydrophilic polymers, and host molecules are substituted; and a physiologically active material linked to a guest molecule, wherein the poly(phosphazene) and the physiologically active material form a conjugate by inclusion of the guest molecule in the host molecule via a host-guest interaction.

Genetically modified non-human mammals and cells
Genetically modified mammals are described which lack the mannan binding lectin associated serine protease MASP-2, together with methods and constructs for their production. Such mammals are useful as models for disorders of the complement system, and in the identification of treatments for such disorders. Also described are mammals which lack the associated protein MAp19; such mammals may also lack MASP-2.
