This disclosure relates to the field of exosome delivery systems. In particular, compositions comprising adipose-derived exosomes that may be used as a delivery system are encompassed. The exosome delivery system can be used to deliver exogenous cargo such as miRNA and other inhibitory RNAs, as well as proteins, to target cells in a subject.

The invention provides methods, cells and constructs for optical measurement of membrane potential. These methods can be used in cells that are not accessible to presently available methods using electrodes. The methods can be directed to, for example, high-throughput drug screening assays to determine agents that can affect membrane potential of a target cell.

AAV MUTANT THAT EFFICIENTLY INFECTS SUPPORTING CELLS

Provided is an AAV mutant that efficiently infects supporting cells. Specifically, provided is a gene expression vector used for treating hearing disorders, and the described expression vector is an AAV mutant. In the AAV mutant, an expression cassette of a therapeutic gene used for treating hearing disorders is inserted or carried, and additionally, the AAV mutant contains a gene sequence that encodes a viral capsid protein mutant.

MODIFIED VIRAL COMPOSITIONS FOR VIRAL TRANSDUCTION

This disclosure provides compositions and methods for delivering a viral composition to cells, e.g., for cell surface receptor-mediated uptake, and enhanced viral transduction. Viral transduction can be achieved via a modified viral composition that includes a moiety that binds to a cell surface receptor ligand. Modified viral compositions and methods for reducing levels or titers of neutralizing antibodies in a subject in need of viral therapy, e.g., gene therapy, are provided. In some embodiments, the modified viral composition includes empty viral particles that bind and internalize neutralizing autoantibodies. Modified viral compositions including empty viral particles can be administered prior to viral therapy. Also provided are pharmaceutical compositions and kits including a modified viral composition.
