Recombinant PIV5 constructs may include PIV5 lacking the conserved C-terminus of the V protein (PIV5V?C), PIV5 with mutations in the N-terminus of the V/P protein (PIV5CPI?), and PIV5 expressing MDA-7/IL-24 (rPIV5-MDA7), rPIV5-V/P-CPI?, rPIV5-CPI+, rPIV5-Rev, rPIV5-RL, rPIV5-P-S157A, rPIV5-P-S308A, rPIV5-L-A1981D, rPIV5-F-5443P, rPIV5-MDA7, rPIV5?SH-CPI?, or rPIV5?SH-Rev. Also included are methods of making and using such oncolytic agents and compositions including such oncolytic agents.

IMMUNOGENIC COMPOSITION AND USES THEREOF
20240197856 · 2024-06-20 ·

Disclosed herein is an immunogenic composition for enhancing immunogenicity and treatment efficacy. The immunogenic composition comprises a nanoparticle (e.g., a nanodiamond) and an immunogenic conjugate having formula (I): X-L-Y, wherein X is an agonist module, L is a linker unit, and Y is an antigen. In the immunogenic composition, the immunogenic conjugate is coupled to the outer surface of the nanoparticle. Also encompasses herein is a method of preventing or treating a disease in a subject in need thereof, comprising administering an effective amount of the immunogenic composition set forth above to the subject.

Compositions and methods comprising hydrocarbon-stapled polypeptides

The invention provides compositions, kits and methods utilizing polypeptides having a viral alpha-helix heptad repeat domain in a stabilized -helical structure (herein also referred to as SAH). The compositions are useful for treating and/or preventing viral infections. The invention is based, at least in part, on the result provided herein demonstrating that viral hydrocarbon stapled alpha helical peptides display excellent proteolytic, acid, and thermal stability, restore the native alpha-helical structure of the peptide, are highly effective in interfering with the viral fusogenic process, and possess superior pharmacokinetic properties compared to the corresponding unmodified peptides.

The invention provides an adenovirus or adenoviral vector characterized by comprising one or more particular nucleic acid sequences or one or more particular amino acid sequences, or portions thereof, pertaining to, for example, an adenoviral pIX protein, DNA polymerase protein, penton protein, hexon protein, and/or fiber protein.

Peptide/?-1,3-glucan complex and production method thereof and pharmaceutical composition containing peptide/?-1,3-glucan complex

This peptide/?-1,3-glucan complex includes ?-1,3-glucan and a peptide/polynucleotide conjugate in which an antigenic peptide is bonded covalently to a polynucleotide or a derivative thereof. The polynucleotide or derivative thereof of the peptide/polynucleotide conjugate bonds via a hydrogen bond with ?-1,3-glucan, forming a complex having a triple helix structure including a single molecular chain of the polynucleotide or derivative thereof and two molecular chains of the ?-1,3-glucan. Alternatively, the side chain of the ?-1,3-glucan and the antigenic peptide are bonded covalently formed by either a cycloaddition reaction between an alkyne and an azide derivative, or a reaction between a maleimide group or a vinyl sulfone group and a thiol group.
