The instant disclosure is generally related to compositions and methods for obtaining and constructing Bacillus licheniformis host cells (e.g., protein production host cells, cell factories) having increased protein production capabilities. Certain embodiments of the disclosure are directed to efficient genetic modifications of B. licheniformis cells and the subsequent selection of such B. licheniformis cells having increased protein production capabilities. Certain other embodiments of the disclosure are generally related to methods and compositions for producing/obtaining auxotrophic B. licheniformis cells, wherein certain other embodiments of the disclosure are directed to methods and compositions for restoring prototrophy in auxotrophic B. licheniformis cells, and expressing genes of interest (GOIs) in such restored prototrophy B. licheniformis cells.
METHODS OF CULTURING PROBIOTIC BACILLI
A method of culturing bacteria of the Bacilli class is disclosed. The method comprises: (a) adding the bacteria to a medium comprising pasteurized starch fibers of a legume of a leguminous plant, and (b) culturing the bacteria under conditions that promote generation of a biofilm of the bacteria on the starch fibers.

Provided is a method for collecting an objective substance such as vanillin from a fermentation broth. Upon collecting an objective substance from a fermentation broth containing the objective substance by solvent extraction using an organic solvent, emulsification during the solvent extraction can be prevented by treating the fermentation broth with a protease and then subjecting it to the solvent extraction, or by carrying out the solvent extraction with an agitation power adjusted to a predetermined range, and thereby the objective substance can be collected from the fermentation broth.

The subject invention provides methods for improving the treatment of effluents and waste matter produced during food processing and production. In particular, the subject invention provides methods for remediating fats, oils and greases (FOG), suspended solids, proteins, and other organic matter that are discharged from plants that process, for example, meats, poultry, seafood, dairy and plant-based oils. The methods of the subject invention utilize a customized microbial cocktail comprising facultative anaerobes, in combination with one or more microbial growth by-products, e.g., enzymes and/or biosurfactants, to digest and/or liquefy food processing waste matter.

The present invention relates to a method for recovering a protein of interest from a bacterial fermentation broth, wherein the bacterial cells in the fermentation broth exhibit a high degree of lysis, comprising the steps of adding to the fermentation broth comprising the protein of interest at least one flocculant in an amount of 0.5 to 50 g/l of the fermentation broth; separating the protein of interest from at least the bacterial cells by filtration, preferably by dead-end filtration or microfiltration; and thereby obtaining the protein of interest in the filtrate; wherein the bacterial cells are Bacillus cells. Further, the invention relates to the use of a flocculant for recovering a protein of interest from a bacterial fermentation broth comprising a high degree of lysed cells.
