Methods for sample preparation and testing

A method and system for analyzing a sample potentially comprising a target, comprising: (i) activating one or more components of the sample analysis system in preparation for a sample, wherein the sample analysis system comprises a heating element, a centrifuge, a thermocycler, an imager, a user interface, and a processor; (ii) obtaining a sample potentially comprising a target; (iii) adding the obtained sample to a sample preparation receptacle, the sample preparation receptacle comprising one or more reagents configured to maximize success of the sample analysis; (iv) heating the sample preparation receptacle, with the added sample, in the heating element of the sample analysis system at a first temperature for a first heating period of time; (v) centrifuging the sample preparation receptacle, after heating, for a first period of time using the centrifuge of the sample analysis system; (vi) transferring at least a portion of the sample, after centrifugation, from the sample preparation receptacle to a sample analysis receptacle, the sample analysis receptacle comprising one or more reagents configured for a qPCR reaction specific to a selected target; (vii) obtaining, via the thermocycler and imager of the sample analysis system, a fluorescence curve during a qPCR reaction; (viii) analyzing, by the process of the sample analysis system, the obtained fluorescence curve to determine a presence or absence of the selected target in the sample; and (ix) reporting, to a user by the user interface of the sample analysis system, the presence or absence of the selected target in the sample; wherein one or more steps of the method are guided by interactive instructions provided by the sample analysis system.

A testing method of thermal barrier coating (TBC) is for evaluating the presence or absence of damage to TBC formed on a bending part on which compression stress acts. The method includes a test piece that includes a pair of arm parts, a bending part arranged between the pair of arm parts, and a TBC layer on a bending surface of the bending part; attaching the test piece to a compression testing device after preparing the test piece; and applying compression stress to the test piece in a direction for bringing the pair of arm parts close together after attaching the test piece with the compression testing device. The pair of arm parts are arranged so as to separate from each other from base end portions toward front end portions of the arm parts. The bending part is arranged between the base end portions.

Extraction reagent of immunosuppressant drug for immunoassays

A reagent for extracting immunosuppressant drugs from a whole blood sample for immunoassay includes protein denaturant, proteolytic enzyme, surfactant and pH buffer. A method and an immunoassay kit for detection of the immunosuppressant concentration in a whole blood sample uses the extraction reagent. The extraction reagent doesn't need the use of organic solvent as that in the traditional extraction methods, therefore the adverse effects of the organic solvent on the antibody activity in a detection system and the other relative defects associated to its use are obviated. The drug extraction process doesn't need centrifugation, as the processed sample can be directly applied for immunoassay. The operation for drug extraction is simple, and the detection result based on this extraction method is accurate.

The system can include a slide sled that can align a smearing slide with a surface of the slide including a fluid sample is disposed, and support the smearing slide at a predefined angle with respect to the surface of the slide. In another example, a method includes obtaining a slide including a sample disposed on a surface, positioning the slide below to a jet nozzle, and dispensing a micro stream of a fluid onto the sample using the jet nozzle.
