The present disclosure provides methods for identifying a patient for varicocele correction. The method includes using the CAP-Score™ Assay to assess whether fertility is affected by the varicocele. The present disclosure also provides methods for identifying a reproductive approach inpatients suffering from varicocele. The method includes using the CAP-Score™ Assay to identify and administer the reproductive approach.

According to some aspects, the present disclosure provides a method and kit for pairing female donor subjects with female recipient subjects, and for selecting female donor subjects. According to other aspects, the present disclosure provides a method of generating a microbiome database.

This invention provides for a method for improving the success rate of in vitro fertilization in women hosting abnormal vaginal microbiota. The method comprises the steps of: (i) selecting a woman suspected of hosting abnormal vaginal microbiota (AVM); (ii) administering to the woman a suitable antibiotic administered in an amount and duration effective to reduce the quantity of abnormal vaginal microbiota hosted by the woman; (iii) administering to the woman via vaginal administration an amount of Lactobacillus species in an amount sufficient to colonize the vaginal mucosa; and, (iv) transferring an embryo to the woman.

A method for sorting motile cells includes introducing an initial population of motile cells into an inlet port of a microfluidic channel, the initial population of motile cells having a first average motility; incubating the population of motile cells in the microfluidic channel; and collecting a sorted population of motile cells at an outlet port of the microfluidic channel. The sorted population of motile cells has a second average motility higher than the first average motility.

Provided herein are methods, systems, and kits for improving success rates in assisted reproductive technologies such as in vitro fertilization, frozen embryo transfer, and intrauterine insemination. These methods, systems, and kits rely on levels of protein, metabolite, and microRNA markers determined herein to be linked to uterine toxicity and embryo implantation failure.

A system and method for sorting sperm is provided. The system includes a housing and a microfluidic system supported by the housing. The system also includes an inlet providing access to the microfluidic system to deliver sperm to the microfluidic system and an outlet providing access to the microfluidic system to harvest sorted sperm from the microfluidic system. The microfluidic system provides a flow path for sperm from the inlet to the outlet and includes at least one channel extending from the inlet to the outlet to allow sperm delivered to the microfluidic system through the inlet to progress along the flow path toward the outlet. The microfluidic system also includes a filter including a first plurality of micropores arranged in the flow path between the inlet and the outlet to cause sperm traveling along the flow path to move against through the filter and gravity to reach the outlet.

Methods for quantifying GDF-9-BMP-15 heterodimers and GDF-9 and BMP-15 homodimers using antibodies directed to epitopes in GDF-9 and BMP-15 in a sandwich ELISA are provided. Also provided are kits for quantifying the heterodimers.

The present Application provides in one aspect a method of diagnosing a menstrual cycle event in a subject (e.g., a WOI), comprising detecting in a biological sample a gene signature for one or more endometrial cell types (e.g., unciliated epithelial cells). The present Application in another aspect provides a method comprising determining a gene expression profile in each of a plurality of endometrial cells, wherein said endometrial cells are: (a) in an endometrial sample obtained from a subject, and (b) unciliated epithelial cells. In still another aspect, the present Application provides a method for detecting that a subject is within a window of implantation (WOI), the method comprising: (a) determining a level of expression of a gene signature in a sample of endometrial cells obtained from a subject, (b) comparing the determined level of expression of each gene in the gene signature with a control level; and (c) determining whether the subject is within the WOI, wherein the subject is identified as being within the WOI if the level of the expression of the gene signature is higher than a control level.
