Clotting factor preparations for delivery into tissue of the intestinal tract using a swallowable drug delivery device

Embodiments provide devices, preparations and methods for delivering therapeutic agents (TAs) such as clotting factors (CFs, e.g., Factor 8) within the GI tract. Many embodiments provide a swallowable device e.g., a capsule for delivering TAs into the intestinal wall (IW). Embodiments also provide TA preparations configured to be contained within the capsule, advanced from the capsule into the IW and/or surrounding tissue (ST) and degrade to release the TA into the bloodstream to produce a therapeutic effect (e.g., improved clotting). The preparation can be operably coupled to delivery means having a first configuration where the preparation is contained in the capsule and a second configuration where the preparation is advanced out of the capsule into the IW or ST (e.g., the peritoneal cavity). Embodiments are particularly useful for delivery of CFs for treatment of clotting disorders (e.g., hemophilia) where such CFs are poorly absorbed and/or degraded within the GI tract.

A disengageable frictional coupling member allows this relative rotation when, for implantation, the tubular body receives an external rotational stress, and that until a predetermined limit torque triggering the disengagement. At explantation, this disengagement is prevented to allow a joint rotation of the tubular body and of the front-end unit and the unscrewing of the helical screw. It is provided for that purpose two conjugated plates facing each other, with flat surfaces such as circular sectors offset in opposite directions with respect to a radial reference plane, in such a way as to form steps providing an anti-disengagement abutment function.

A motor is also disposed within the interior chamber and is configured to rotate one of the cap and the one or more sample collection chambers so to align one or the one or more sample collection chambers and the sampling port of the cap so to allow for sample collection. A microcontroller is also disposed within the interior chamber and is in communication with at least the motor. The microcontroller is configured to control the selective alignment of the sampling port and one of the one or more sample collection chambers and induce gastric intestinal fluid sampling.
