ORALLY BIOAVAILABLE LIPID-BASED CONSTRUCTS
The present invention is embodied by a composition capable of chaperoning a typically non-orally available therapeutic or diagnostic agent through the environment of the digestive tract such that the therapeutic or diagnostic agent is bioavailable. The composition may or may not be targeted to specific cellular receptors, such as hepatocytes. Therapeutic agents include, but are not limited to, insulin, calcitonin, serotonin, and other proteins. Targeting is accomplished with biotin or metal based targeting agents.

Methods for preparing micro-particles using a double emulsion technique combining a membrane and a micro-sieve are provided. Particularly the present invention relates to method for preparing micro-particles comprising: preparing a first phase comprising an active agent; preparing a second phase comprising a carrier and a solvent; passing the first phase and the second phase through a membrane to form a primary emulsion; passing the primary emulsion through a micro-sieve in a continuous phase to form a secondary emulsion; and removing the solvent to form the micro-particles.

Unlike existing probes that are cleared within a few days in vivo, the probes described herein (e.g., OFS-1) allow for monitoring resorption over the course of longer time periods with a single dose.
