A method of monitoring the level of an analyte such as glucose in a subject comprises repeated steps of extracting a quantity of the analyte from the subject into a sample, for example using reverse iontophoresis, then measuring the concentration of the analyte with a method that depletes the analyte. Whereas known methods aim to deplete the analyte fully between extraction steps, the present method allows the concentration to build up with each cycle and measures successive changes in concentration. The higher concentrations present permit more reliable measurements to be made. The sample may be primed with an initial quantity of the analyte.

A means and a method is disclosed to diminish or eliminate the pain associated with a sharp object penetrating the skin, during such procedures as an injection, biopsy, or deriving a blood sample. To this end, repeated tapping, pressing, or rubbing or vibrating is performed over the skin at or near the site of penetration of the sharp object in conjunction with applying electricity on the skin. The invention discloses a method of using a skin-puncturing means, with enhanced features, to provide local anesthesia at the site of penetration of a sharp object.

Microporation is accomplished to form a micropore of selected depth in the biological membrane and the porated site is contacted with the permeant. Additional permeation enhancement measures may be applied to the site to enhance both the flux rate of the permeant into the organism through the micropores as well as into targeted tissues within the organism.
