TUNEABLE DELIVERY OF NANOPARTICLE BOUND ACTIVE PLASMIN FOR THE TREATMENT OF THROMBOSIS
Compositions and methods for therapeutic delivery are disclosed. More particularly, the present disclosure relates to nanoparticle compositions that sequester the activity of a target molecule while leaving other domains accessible to bind targeted tissues of interest. Methods for thrombus dissolution include administering a nanoparticle reversibly coupled to a target molecule that can dissolve a blood clot. Compositions and methods for inducing blood clotting are also disclosed. Methods for inducing blood clotting include administering a nanoparticle reversibly coupled to a target molecule that can induce the formation of a blood clot. Methods for sequestering a target molecule are also disclosed. The method includes reversibly coupling a target molecule to a nanoparticle having an affinity ligand that reversibly couples the target molecule, and thus, sequesters the target molecule activity until the target molecule interacts with its substrate resulting in the release of the target molecule.

The present invention is directed to hemostatic compositions comprising at least partially integrated agglomerated ORC fibers, fibrinogen, and thrombin and methods of forming a powdered hemostatic composition, comprising the steps of: forming a suspension of a mixture comprising particles of fibrinogen, thrombin, ORC fibers in a non-aqueous low boiling solvent, agitating and shearing said suspension in a high shear mixing reactor, adding water to allow particles to agglomerate, allowing the non-aqueous solvent to evaporate, drying and sieving the composition; and thus forming the powdered hemostatic composition.

The present disclosure further discloses a method for delivering osteoblast cells into a subject. The method for obtaining osteoblast cell-mixture as disclosed herein is devoid of any additives like calcium chloride and aprotinin. The method for delivering osteoblast cells as disclosed herein provides bone regeneration in the subject.

The present invention provides a composition suitable as a composition for treating blood coagulation and/or complement disorders (raw materials for producing therapeutic agents for these diseases) and a method for effectively producing the composition. A method for producing a composition for treating blood coagulation and/or complement disorders according to the present invention includes the following steps of: (1) embedding an organoid formed from vascular endothelial cells or vascular endothelial cells and hepatocytes, in an extracellular matrix; (2) culturing the extracellular matrix; and (3) collecting a culture supernatant from the culture obtained in the step (2).
