The present disclosure relates to polynucleotides comprising an open reading frame of linked nucleosides encoding human interleukin-12 (IL12), functional fragments thereof, and fusion proteins comprising IL12. In some embodiments, the open reading frame is sequence-optimized. In particular embodiments, the disclosure provides sequence-optimized polynucleotides comprising nucleotides encoding the polypeptide sequence of human IL12, or sequences having high sequence identity with those sequence optimized polynucleotides.

The present invention provides a pluripotent stem cell or a derivative thereof. A first nucleic acid molecule is introduced into a genome of the pluripotent stem cell or the derivative thereof; and a second nucleic acid molecule is introduced to a 3′UTR region of an immune response-related gene in the pluripotent stem cell or the derivative thereof. The first nucleic acid molecule encodes a small nucleic acid molecule that mediates RNA interference, and the small nucleic acid molecule can specifically bind to a transcript product of the second nucleic acid molecule to start an RNA interference program to degrade or silence mRNA of the immune response-related gene, thereby blocking the expression of the immune response-related gene, such that the cell has immunological compatibility, and thus can eliminate or reduce alloimmune rejection responses.

The circular RNAs can comprise group I intron fragments, spacers, an IRES, duplex forming regions, and/or an expression sequence, thereby having the features of improved expression, functional stability, low immunogenicity, ease of manufacturing, and/or extended half-life compared to linear RNA. Pharmaceutical compositions comprising such circular RNAs and transfer vehicles are particularly suitable for efficient protein expression in immune cells in vivo. Also disclosed are precursor RNAs and materials useful in producing the precursor or circular RNAs, which have improved circularization efficiency and/or are compatible with effective circular RNA purification methods.
