Vortex generator for agitation of fluids during sample preparation

An apparatus, vortex generator assembly and method for automated cell lysis and nucleic acid purification and processing. The vortex generator assembly includes sample holder having a lysis well, at least one wash well, and an elution well. The vortex generator assembly also includes a sample holder cover having a plurality of vibration rods for creating a vortex in the wells of the sample holder. The apparatus includes motor operating a rotating cam to cause the vibration rods to vibrate and create the vortex in a well holding fluid and magnetic beads, wherein the vortexing speed is sufficient to overcome the magnetic attraction between the beads and disperse the beads in solution, to collect nucleic acids such as DNA.

BIOSENSOR FOR SINGLE CELL ANALYSIS

A biosensor for single cell analysis is disclosed. The biosensor includes a substrate, an array of electrodes, and a passivation layer. The substrate includes a roughened surface, where the array of electrodes is patterned on the roughened surface. Each electrode includes a distal tip and a proximal end. The passivation layer is deposited on top of the biosensor and includes a microwell around the distal tip of an electrode. A single cell is trapped within the microwell and adhered onto the distal tip of the electrode for further single cell analysis.

An exemplary embodiment of the present disclosure provides a system for detecting antimicrobial resistance of a bacteria in a biological sample. In some embodiments, the system can include a plurality of containers, a detecting agent in each of the plurality of containers, and an antimicrobial agent in at least a portion of the plurality of containers. The antimicrobial agent is disposed in at least one of the plurality of containers. Each of the containers can contain at least a portion of the biological sample. The detecting agent can be configured to produce optically detectable changes responsive to bacterial respiration or growth, directly from patient samples of from patient sample cultures.

For the multiwell plates, the bottom of at least one well has a layer of porous matrix disposed thereon, or is comprised of a layer of porous matrix. The layer of porous matrix permits a low-volume liquid sample to distribute evenly across the porous matrix. Also disclosed herein are methods of performing a photometric or spectrophotometric measurement on a liquid sample having a small volume, by using a multiwell plate comprising a layer of porous matrix.
