Methods and systems for aligning dispensing arrays with microfluidic sample arrays
A method of registering a location of a dispenser array in relation to a microfluidic array is provided. One of the dispenser array and the microfluidic array can be movable in relation to the frame, and the other can be fixed relative to the frame. Their relative positions can be identified by a set of coordinates. Identification of a fiducial marker can occur in a manner permitting the fiducial reference to appear in a first position of a field of view of a first camera when the dispenser array or the microfluidic array is in an alignment position. Quantities related to a vector displacement from the alignment position to a fixed position on the microfluidic array or the dispenser array can be identified. Quantities determined can be used to guide positioning of the dispenser array relative to the microfluidic array.

An apparatus suitable for single molecule sequencing. The apparatus includes at least one nanowell, a plurality of nucleic acid immobilization moieties, and a plurality of types of nucleic acid fragments. The nanowell has an observation zone. The nucleic acid immobilization moieties are disposed in or proximate to the observation zone. The nucleic acid fragments are immobilized to the nucleic acid immobilization moieties, respectively. At least one polymerase is disposed in the observation zone. A method of sequencing nucleic acid molecules using the above-mentioned apparatus is provided.

The invention is directed to a device and a method for carrying out experiments of chemical, biochemical, biological and physical analysis, reaction and assay employing a reaction unit, in a preferred embodiment comprising: a capillarity reaction chamber, non-capillarity zone and bottom structure, which can use capillary action to quantitatively take up by itself and hold liquid in the reaction chamber. A multi-unit plate, which contains a number of reaction units on a plate frame, can be used for parallel experiments in conjunction with reservoir plate, waster pad, and liquid transfer guider.

A cover assembly may include a tray assembly frame, a first cover supported by the tray assembly frame, the first cover extending in a first plane and defining one or more first openings; a second cover supported by the tray assembly frame, the second cover extending in a second plane and defining one or more second openings, wherein the first and second planes are different planes, and wherein the second cover is disposed above the first cover. Provided herein is a system comprising: a) spatial sampler system configured to collect and transmit one or a plurality of cells or nuclei from a tissue specimen, the system comprising: i) a lower carrier having an array of conduits passing therethrough, each conduit comprising an opening on a first side and communicating with an opening on a second side, wherein the opening on the second side either (1) terminates in a capillary or (2) opens onto a well of a multiwell plate; ii) positioned, above the lower carrier, a perforated specimen holder configured to support a frozen tissue specimen, wherein the specimen holder comprises a plurality of perforations having a size sufficient to permit the passage of single cells or nuclei; and iii) positioned, above the specimen holder, a multifunctional head comprising an upper array of upper conduits, optionally, each aligned with a conduit opening of the lower carrier.
Nanoliter array loading
An interface is provided for storing microfluidic samples in a nanoliter sample chip. A fluid access structure provides a fluid access region to a selected subset of sample wells from an array of sample wells. A fluid introduction mechanism introduces a sample fluid to the fluid access region so that the sample wells in the selected subset are populated with the sample fluid without the unselected sample wells being populated with the sample fluid.
Apparatus and Methods for Parallel Processing of Microvolume Liquid Reactions
Disclosed herein are apparatuses and methods for conducting multiple simultaneous micro-volume chemical and biochemical reactions in an array format. In one embodiment, the format comprises an array of microholes in a substrate. Besides serving as an ordered array of sample chambers allowing the performance of multiple parallel reactions, the arrays can be used for reagent storage and transfer, library display, reagent synthesis, assembly of multiple identical reactions, dilution and desalting. Use of the arrays facilitates optical analysis of reactions, and allows optical analysis to be conducted in real time. Included within the invention are kits comprising a microhole apparatus and a reaction component of the method(s) to be carried out in the apparatus.

In some aspects, methods of loading a molecule of interest into a sample well involve loading a molecule of interest into a sample well in the presence of a crowding agent and/or a condensing agent. In some aspects, methods of loading a sequencing template into a sample well are provided.
