FISH EGG PROCESSING APPARATUS
There is provided a fish egg processing apparatus capable of performing a predetermined processing on fertilized eggs with higher efficiency. A gene injection device 1 which is a fish egg processing apparatus according to the present invention includes a processing water tank 8 which processes collected fish eggs e and a processing device for injecting a predetermined substance into the fish eggs e positioned in the processing water tank 8, wherein the processing water tank 8 includes: a region prior to processing 81 where the width dimension is set such that a plurality of the fish eggs e are not allowed to lie side by side and form a line in order to house the fish eggs e prior to processing; and a region after processing 82 for housing the fish eggs e on which a predetermined processing has been performed.
Development of YY male fish broodstocks in a single generation
This disclosure describes exemplary embodiments of a method of creating a YY animal broodstock, preferably in a single generation, wherein the broodstock includes only sperm-producing YY males and egg-producing YY males, the method comprising the steps of: (a) creating YY males via androgenesis; (b) exposing selected ones of the YY males created in step (a) to a feminizing hormone; and (c) identifying sperm-producing YY males and egg-producing YY males from among the YY males created in steps (a) and (b). In other embodiments, the method further comprises: (d) repeating steps (a) through (c) N times in order to produce N unrelated families of sperm-producing YY males and egg-producing YY males; and (e) cross-breeding various ones of the unrelated families produced in step (d) in order to produce a genetically-diverse YY progeny. In some embodiments, N may be about 60.

METHOD OF PREPARING ddx27-DELETION ZEBRAFISH MUTANTS
A method of preparing a ddx27-deletion zebrafish mutant, including: determining a target of ddx27 knockout on a sixth exon of the ddx27 in a zebrafish and designing a gRNA sequence; using primers T7-ddx27-sfd and tracr rev for PCR amplification with a pUC19-gRNA scaffold plasmid as a template; purifying and transcribing the PCR product obtained in vitro to produce gRNA; introducing the gRNA and a Cas9 protein into the zebrafish; and culturing the zebrafish to obtain a zebrafish ddx27 mutant of stable inheritance. In addition, the application also discloses a phenotype of the ddx27-deletion zebrafish mutant, which plays an important role in investigating the biological function.
Immune-compromised zebrafish
Genetically-modified zebrafish lacking one or more immune-related genes, and the use thereof, e.g., in cell or tissue transplantation methods or in stem cell biology. In a first aspect, the invention provides a genetically-modified fish whose genome is homozygous for engineered or induced genetic alteration, e.g., an alteration that changes the sequence of the genomic ONA resulting in insertion and deletion of nucleotides that disrupt protein function or shift the frame of translation leading to premature protein termination, in one or more immune-related genes selected from the group consisting of: foxn1, rag2, jak3, prkdc, and interleukin 2-receptor gamma a and (IL2RGa and IL2RGb), wherein the genetic alteration results in an inactivation (i.e., loss of expression or function) of both alleles of the immune-related gene.

Some of the embodiments of the invention provide for making a founder animal that is completely free of all unplanned genetic modifications. Some embodiments are directed to removing genetic faults in established breeds without making other alterations to the genome. Other embodiments are directed to particular tools or processes such as TALENs or CRISPR with a preferred truncation. One embodiment involves introducing a targeted targeting endonuclease system and a HDR template into a cell (optionally with a mismatch in the binding of the targeting endonuclease and the targeted site). Another embodiment includes processes of making a genetically modified livestock animal comprising a genome that comprises inactivation of a neuroendocrine gene selective for sexual maturation, with the inactivation of the gene preventing the animal from becoming sexually mature. One embodiment includes compositions and methods for making livestock with a polled allele, including migrating a polled allele into a bovine species without changing other genes or chromosomal portions.

LABORATORY ZEBRAFISH AUTISM BEHAVIORAL MODEL APPARATUS
A laboratory zebrafish autism behavioral model apparatus is provided. The laboratory zebrafish autism behavioral model apparatus comprises a model body, wherein the model body is divided into a plurality of experimental units, and each of the experimental units has a cuboid structure with completely identical shapes; a free swimming area, a stimulus area, and a normal non-stimulus area are formed on each of the experimental units; the stimulus area and the normal non-stimulus area are located on two sides of the free swimming area, respectively, and the free swimming area, the stimulus area, and the normal non-stimulus area are interconnected via mesh openings. The zebrafish autism behavioral model apparatus facilitates further exploration of autism etiology, effectively resolving interference between the stimulus area and the zebrafish's innate thigmotactic behavior, reducing experimental errors, saving time, and minimizing instrument wear and costs.
