20210259219 · 2021-08-26 ·

The present invention generally relates to genetically modified swine wherein at least one allele of a SIGLEC1 gene has been inactivated and/or at least one allele of a CD163 gene has been inactivated. Genetically modified swine having both alleles of the SIGLEC1 gene and/or both alleles CD163 gene inactivated are resistant to porcine reproductive and respiratory syndrome virus (PRRSV). Methods for producing such transgenic swine are also provided.

Humanized IL-6 and IL-6 receptor

Mice that comprise a replacement of endogenous mouse IL-6 and/or IL-6 receptor genes are described, and methods for making and using the mice. Mice comprising a replacement at an endogenous IL-6Rα locus of mouse ectodomain-encoding sequence with human ectodomain-encoding sequence is provided. Mice comprising a human IL-6 gene under control of mouse IL-6 regulatory elements is also provided, including mice that have a replacement of mouse IL-6-encoding sequence with human IL-6-encoding sequence at an endogenous mouse IL-6 locus.

GENETICALLY-MODIFIED POULTRY EGG

Provided are a poultry knock-in egg and knock-out egg. The present invention pertains to a knock-out poultry egg in which at least one oviduct-specific gene has been knocked out, said gene being selected from the group consisting of ovalbumin, ovomucoid, ovomucin, ovotransferrin, ovoinhibitor, and lysozyme, and at least one egg allergen protein has been reduced or eliminated, said protein being selected from the group consisting of ovalbumin, ovomucoid, ovomucin, ovotransferrin, ovoinhibitor, and lysozyme.

A METHOD OF GENERATING STERILE AND MONOSEX PROGENY

The disclosure provides a method of generating a sterile sex-determined fish, crustacean, or mollusk. The method comprises breeding (i) a fertile homozygous mutated female fish, crustacean, or mollusk having at least a first mutation and a second mutation with (ii) a fertile homozygous mutated male fish, crustacean, or mollusk having at least the first mutation and the second mutation to produce the sterile sex-determined fish, crustacean, or mollusk. The first mutation disrupts one or more genes that specify sexual differentiation, the second mutation disrupts one or more genes that specify gamete function, and the fertility of the fertile homozygous female fish, crustacean, or mollusk and the fertile homozygous mutated male fish, crustacean, or mollusk has been rescued. The disclosure also provides methods of making broodstock for use in producing sterile sex-determined fish, crustacean, or mollusks, as well as the broodstock itself.

Mutant Mouse-Derived Pancreatic Organoid and Method for Evaluating Standardized Drug for Efficacy
20210282378 · 2021-09-16 ·

Provided are three-dimensional pancreatic organoids derived from the pancreas of a genetically mutated mouse, a method for producing the three-dimensional pancreatic organoids, the use of the three-dimensional pancreatic organoids for drug effect verification and/or drug screening, and a universally applicable standardized drug effect evaluation method/drug screening method.

The present invention provides ungulate animals, tissue and organs as well as cells and cell lines derived from such animals, tissue and organs, which lack expression of functional endogenous immunoglobulin loci. The present invention also provides ungulate animals, tissue and organs as well as cells and cell lines derived from such animals, tissue and organs, which express xenogenous, such as human, immunoglobulin loci. The present invention further provides ungulate, such as porcine genomic DNA sequence of porcine heavy and light chain immunogobulins. Such animals, tissues, organs and cells can be used in research and medical therapy. In addition, methods are provided to prepare such animals, organs, tissues, and cells.

METHODS FOR GENERATING ANIMALS WITH DESIRABLE TRAITS

Method for selecting mammalian cells having a genetically-desirable trait, the method comprising culturing an embryo in vitro, dividing the cells from the embryo into aliquots, subjecting the cells from at least one of the aliquots to a genetic analysis, and, based on the results of such analysis, selecting an aliquot of cells. Method for selecting mammalian cells having a genetically-desirable trait, the method comprising culturing an embryo in vitro, transferring the cultured embryo into a recipient female, collecting the embryo, dividing the cells from the embryo into aliquots, subjecting the cells from at least one of the aliquots to a genetic analysis, and, based on the results of such analysis, selecting an aliquot of cells. Method for selecting a mammalian embryo having a genetically-desirable trait, the method comprising removing one or more cells from each of a plurality of embryos, culturing the cells, subjecting the cultured cells to a genetic analysis, and, based on the results of such analysis, selecting an embryo.

NON-MEIOTIC ALLELE INTROGRESSION

Methods, uses, and compositions for manipulating genomic DNA. Some of the embodiments of the invention provide for making a founder animal that is completely free of all unplanned genetic modifications. Some embodiments are directed to removing genetic faults in established breeds without making other alterations to the genome. Other embodiments are directed to particular tools or processes such as TALENs or CRISPR with a preferred truncation. One embodiment involves introducing a targeted targeting endonuclease system and a HDR template into a cell (optionally with a mismatch in the binding of the targeting endonuclease and the targeted site). Another embodiment includes processes of making a genetically modified livestock animal comprising a genome that comprises inactivation of a neuroendocrine gene selective for sexual maturation, with the inactivation of the gene preventing the animal from becoming sexually mature. One embodiment includes compositions and methods for making livestock with a polled allele, including migrating a polled allele into a bovine species without changing other genes or chromosomal portions.
