CELL CULTURING SYSTEM AND METHOD

A cell culturing system (100) comprises a docking station (104, 105), a handling unit (102), a culturing module (101) and an actuation layer. The culturing module has a culturing well and a culturing membrane separating the culturing well in an apical culturing chamber and a basal culturing chamber. The handling unit has a seat for removably accommodating the culturing module and the actuation layer and a bottom with an actuation bore, wherein, when the culturing module is arranged in the seat, the actuation bore is associated to the culturing well and the bottom is separated from the culturing module by the actuation layer. The docking station has a coupling structure for removably holding the handling unit in a predefined position and an actuation feeding channel, wherein, when the handling unit is held by the coupling structure in the predefined position, a first end of the actuation feeding channel is connected to the actuation bore and a second end of the actuation feeding channel is connected to a connector.

Compositions of matter, methods, modules, and automated instruments may relate to synthesizing a library including an editing cassette including a different gRNA and donor DNA pair, amplifying the editing cassette in a partition separate from other editing cassettes in the library, adding nuclease to the partition, and adding lipofectamine to the editing cassette and nuclease to form a lipofectamine/nucleic acid/nuclease complex. A microcarrier coated in extracellular matrix or a cell adhesion molecule coating may be added to the lipofectamine/nucleic acid/nuclease complex. Cell growth material, the microcarrier, and mammalian cells may be transferred to a growth module in an automated closed cell editing instrument via a liquid handling system. The mammalian cells may be allowed to seed on the microcarrier. Conditions may be provided for the mammalian cells to take-up and be edited by a payload associated with the lipofectamine/nucleic acid/nuclease complex. The mammalian cells may be detached from the microcarrier.

The system may include a bioreactor device for producing the tissue specimen which includes a packaged/sealed seeding cup containing a scaffold in a sterile environment amenable to seeding with cells. After seeding, the cup may be stored in a standard laboratory culture tray for incubation. The produced tissue specimen may next be transferred to a testing instrument under control of a microcontroller which automatically performs a series of tests on the tissue and generates testing data communicated to an external electronic device which can operably interact with and control operation in part of the testing instrument via software. Tissue integrity in the instrument may be maintained by sterile fluid circulation and heating systems. The system advantageously provides efficient and reproducible tissue growth and analysis.

A cell processing system, fluidics cartridge, and methods for using actuated surface-attached posts for processing cells are disclosed. Particularly, the cell processing system includes a fluidics cartridge and a control instrument. The fluidics cartridge includes a cell processing chamber that has a micropost array therein, a sample reservoir and a wash reservoir that supply the cell processing chamber, and a waste reservoir and an eluent reservoir at the output of the cell processing chamber. A micropost actuation mechanism and a cell counting mechanism are provided in close proximity to the cell processing chamber. A method is provided of using the cell processing system to collect, wash, and recover cells. Another method is provided of using the cell processing system to collect, wash, count, and recover cells at a predetermined cell density.
