A rigid cartridge for housing and removably coupling with a soft body bioreactor is provided. The rigid cartridge includes rigid walls defining a rigid housing. The rigid walls expose a fixed interior configured to house the soft body. An interior surface of a first rigid wall includes a first mating mechanism to engage a second mating mechanism of the soft body to removably coupled with the rigid cartridge and accommodate the soft body in the fixed interior of the rigid cartridge. A rigid wall includes a planar upper surface including an aperture. Each aperture is encompassed by an annular hood protruding upwardly from the substantially planar upper surface and integrally formed with the at least one rigid wall. The aperture is configured to receive and fixed engage a corresponding connector. Each connector includes a rigid coupling mechanism that provides communication with a corresponding plurality of ports of the soft body.

MULTIWELL PLATE WITH INTEGRATED STIRRING MECHANISM

This invention describes a design for a multiwell plate that contains integrated pumps that are used to stir each well of the plate. The device employs microfluidic logic technology to drive each peristaltic pump. This enables the plates to run autonomously, requiring only a static vacuum supply for power. The devices are entirely constructed out of low-cost polymers, with no electronics, and yet contains simple digital logic circuits to control the pumps. A stack of these plates may be run continuously in a standard cell culture incubator, allowing high-throughput culture of organoids.

Embodiments of the invention relate to a cell culture incubator having a gas flow regulation system that exerts control over atmospheric parameters within the incubator. Particular embodiments include an enclosed environmental chamber and a control unit operably linked thereto, the control unit having an oxygen module and a pressure module. Control unit embodiments, by way of these modules, are configured to regulate both oxygen partial pressure and total gas pressure within the enclosed environmental chamber. Embodiments of the control unit are adapted (a) to provide instructions to the oxygen module to regulate an oxygen level to an instructed hypoxic oxygen level and (b) to provide instructions to the pressure module to regulate total gas pressure to an instructed positive pressure level. The regulation of oxygen to the instructed hypoxic level prevails despite an oxygen partial pressure-increasing effect of the positive pressure condition associated with the instructed positive pressure level.

This allows for the measurement and correction of the environmental conditions within the chamber, and the return of the sample to the main chamber. Utilizing this sampling method, the separable bypass flow monitoring chamber may be isolated from the main chamber by a set of valves during a period of high-heat decontamination of the incubator therein protecting the sensors and contents of this separable chamber, as well as allowing the use of less expensive and lower temperature rated incubator sensors.

The electronic controller may be controlled by a host system, such a server. The electronic controller may be configured to direct production of the bioplastic in the process chamber using hydrocolloid, which is derived from algae.
