Fluidic devices are provided and/or configured to form and support, extractable in-situ-formed hydrogels or hydrogel membranes that reside in a hydrogel chamber formed above, and in direct fluid communication with, an underlying fluidic channel, in the absence of an intervening membrane. In some example embodiments, the integrated fluidic device may include a geometrical hydrogel retention structure that provides a restoring force to the hydrogel when fluidic pressure is applied to the hydrogel from the underlying fluidic channel, or a geometrical meniscus-pinning feature that resists flow of a hydrogel precursor solution out of the hydrogel chamber, facilitating the formation of a hydrogel membrane extending over the integrated fluidic channel. The hydrogel or hydrogel membrane may be seeded with cells by delivering a cell-containing liquid to the fluidic channel, optionally while contacting the hydrogel with media provided in a media reservoir residing above the hydrogel layer.

MONITORING, SIMULATION AND CONTROL OF BIOPROCESSES

Methods for monitoring, controlling and simulating a bioprocess comprising a cell culture in a bioreactor are provided. The methods comprise obtaining values of one or more process conditions for the bioprocess at one or more maturities, and determining the specific transport rates of one or more metabolites in the cell culture using the values obtained as input to a machine learning model trained to predict the specific transport rates of the one or more metabolites at a latest maturity of the one or more maturities or a later maturity based at least in part on the values of one or more process conditions for the bioprocess at the one or more preceding maturities. The methods further comprise predicting one or more features of the bioprocess based at least in part on the determined specific transport rates. Systems, computer readable media and methods for providing tools to implement such methods are also provided.

ALIGNMENT AND FUSION OF CELLS WITH AN ELECTRICAL FIELD

A method of alignment and fusion of cells with an electrical field includes firing a first fluid dispenser of an electrofusion device until a first impedance sensor in a first microfluidic channel of the electrofusion device detects a presence of a first cell. The method includes firing a second fluid dispenser of the electrofusion device until a second impedance sensor in a second microfluidic channel of the electrofusion device detects a presence of a second cell. The method includes moving the first cell and the second cell into a merging chamber of the electrofusion device by firing a third fluid dispenser of the electrofusion device, in response to alignment of the first cell in the first microfluidic channel with the second cell in the second microfluidic channel. The method further includes fusing the first cell and the second cell in the merging chamber by creating an electrical field in the merging chamber.

Aspects of the present invention relate to a networked cell culture incubator and to methods for operating such an incubator. In one aspect, the cell culture incubator includes a network interface for communicating with a source of parameter data utilized successfully by other incubators. The incubator receives appropriate parameter data and conducts the incubation process as prescribed by the parameter data so as to provide an improved environment for cell culture growth. The incubator may share its own parameter data with the data source for use by other incubators. The incubator and data source may share other forms of data as well.

Embodiments described herein generally provide for the expansion of cells in a cell expansion system using an active promotion of a coating agent(s) to a cell growth surface in some embodiments. A coating agent may be applied to a surface, such as the cell growth surface of a hollow fiber in a bioreactor, by controlling the movement of a fluid in which a coating agent is suspended, by changing flow rates, by changing flow directions, by rotation of the bioreactor, and/or combinations thereof.

A plurality of multivariate techniques can be used to construct the predictive model capable of forecasting concentrations over multiple and diverse cell lines. The predictive model is also scalable. In one embodiment, a future lactate concentration trajectory is determined and at least one condition within a bioreactor is changed or modified to maintain lactate concentration within desired ranges.
