The present invention relates to the in vitro production of erythrocytes/granulocytes and to the treatment of myelodysplastic syndrome using a method for inducing hemoblast differentiation. The present invention provides a media composition comprising gelsolin as an active ingredient for inducing the differentiation of hematopoietic precursor cells into erythrocytes/granulocytes, and a pharmaceutical composition comprising gelsolin as an active ingredient for treating myelodysplastic syndrome. Since the composition of the present invention improves the efficiency of differentiation of hematopoietic precursor cells into erythrocytes/granulocytes while maintaining a low rate of occurrence of cell dysplasia and having the effect of improving the enucleation rate and cell survivability, the present invention can be effectively used for producing erythrocytes/granulocytes in vitro and for treating myelodysplastic syndrome.

METHODS FOR ISOLATING AND CULTURING TUMOR CELLS
Methods of generating a population of tumor cells, such as circulating tumor cells (CTCs) isolated from fluid from a subject. The methods involve collecting a fluid sample containing the tumor cells from the subject, and culturing the tumor cells in the fluid sample in a three-dimensional cell culture, wherein the three-dimensional cell culture comprises at least one inhibitor of Rho-kinase to generate the population of CTCs. If the fluid is whole blood or contains blood, the method may also involve subjecting the fluid sample to density gradient separation to separate the tumor cells from the fluid prior to culturing. In addition, methods of identifying a candidate treatment for a subject having a condition marked by the presence of tumor cells, methods of monitoring in a subject the persistence, regression, or progression of a disease or condition marked by the presence of tumor cells, and methods of generating a cell line of tumor cells.

The present invention provides a method for producing neural cells or a neural tissue, including the following steps (1)-(3): (1) a first step of culturing pluripotent stem cells in the absence of feeder cells and in a medium containing 1) a TGFβ family signal transduction pathway inhibiting substance and/or a Sonic hedgehog signal transduction pathway activating substance, and 2) a factor for maintaining undifferentiated state, (2) a second step of culturing the cells obtained in the first step in suspension to form a cell aggregate, and (3) a third step of culturing the aggregate obtained in the second step in suspension in the presence or absence of a differentiation-inducing factor to obtain an aggregate containing neural cells or a neural tissue.
MEDIUM COMPOSITION FOR CULTURING STEM CELLS
The present disclosure relates to a medium composition for culturing stem cells, and more specifically, to a medium composition for culturing mesenchymal stem cells, in which the medium composition includes a basic medium in which various quasi-completed mediums (DMEM, α-MEM, IMDM, F12, and DMEM/F12) are mixed, L-ascorbic acid 2-phosphate, fetal bovine serum, basic fibroblast growth factors (b-FGF), insulin, N-acetyl-L-cysteine, calcium chloride, and hydrocortisone.
According to the present disclosure, it is capable of improving proliferation ability and differentiation ability of the mesenchymal stem cells, and is capable of producing cell therapy products more economically using the mesenchymal stem cells by enabling the mesenchymal stem cells to be cultured at a low price compared to the existing culturing methods.
