The present invention provides, among other things, methods of treating phenylketonuria (PKU), including administering to a subject in need of treatment a composition comprising an mRNA encoding phenylalanine hydroxylase (PAH) at an effective dose and an administration interval such that at least one symptom or feature of PKU is reduced in intensity, severity, or frequency or has delayed in onset. In some embodiments, the mRNA is encapsulated in a liposome comprising one or more cationic lipids, one or more non-cationic lipids, one or more cholesterol-based lipids and one or more PEG-modified lipids.

METHOD OF PREPARING SILANOLS WITH SELECTIVE CYTOCHROME P450 VARIANTS AND RELATED COMPOUNDS AND COMPOSITIONS
This disclosure provides a method of preparing a silanol-functional organosilicon compound with a cytochrome P450 variant that facilitates the oxidization of a silyl hydride group to a silanol group in the presence of oxygen. The method includes combining the cytochrome P450 variant and an organosilicon compound having at least one silicon-bonded hydrogen atom to give a reaction mixture and exposing the reaction mixture to oxygen to oxidize the organosilicon compound, thereby preparing the silanol-functional organosilicon compound. Cytochrome P450 variants suitable for use in the method are also disclosed, along with methods for engineering and optimizing the same. Nucleic acids encoding the cytochrome P450 variants and compositions, expression vectors, and host cells including the same are also disclosed.

The present invention also provides strategies, methods, systems, kits and combinations for scalable expression, purification and enhanced production of the virus like proteins of SARS-CoV while maintaining their size range and composition. Such multi-subunit VLPs can be utilized to make immunogenic compositions or vaccines.

Provided are novel flavone hydroxylases, a microorganism for synthesizing flavone C-glycoside compounds, and use thereof. The present inventor obtains novel flavones hydroxylates PhF2H and PhF3′H by cloning, which belong to cytochrome P450 hydroxylates and have the function of hydroxylating specific positions of compounds. Further, the present inventor, by modifying the enzymes and combining a C-glycoside glycosyltransferase and assembly of a synthesis pathway of a flavone precursor, efficiently synthesizes flavone C-glycoside compounds such as oriention, isooriention, vitexin and isovitexin, and related intermediates such as eriodictyol and 2-hydroxynaringenin in the synthesis pathway thereof in an artificial recombinant expression system.
Methods and compositions involving promoters derived from Yarrowia lipolytica
The current methods and compositions provide for nucleotide sequences of promoters from Yarrowia lipolytica which may be used to drive gene expression in a cell. In some aspects, the promoters are useful for modulating lipid production in oleaginous organisms such as yeast.
