Method of culturing NK cells and kits containing medium additions
11613730 · 2023-03-28 · ·

The present invention relates to a method for culturing natural killer cells (NK cells) applied to immune therapy and, more particularly, to a medium addition kin for culturing NK cells with which lymphocytes derived from human peripheral blood can be cultured to effectively amplify and activate NK cells, which have an outstanding therapeutic effect on malignant tumors, while remarkably increasing the share of NK cells, and an NK cell culturing method using the kit.

Indole regulation of antigen presenting cells

The disclosure provides methods and compositions for affecting the development of antigen presenting cell (APC, e.g., a macrophage or dendritic cell). The methods include maturing an APC, promoting anti-inflammatory phenotype, promoting development of a T regulatory cell (Treg) from a naive T cell. The methods generally include exposing an APC to a tryptophan derived microbiota metabolite (TDMM), such as an anti-inflammatory or pro-mucosal TDMM, and permitting the APC to mature. In some embodiments, the conditioned APC is exposed to a naive T cell to further promote development of a T regulatory cell (Treg). In some embodiments, the TDMM is selected from the group consisting of indole, indole-3-acetate, 5-hydroxyindole, and indole-3-pyruvate.

Provided herein are methods and customized media compositions for culturing CIK NKT cells.

EXPANSION OF TUMOR INFILTRATING LYMPHOCYTES FROM LIQUID TUMORS AND THERAPEUTIC USES THEREOF
20230092130 · 2023-03-23 ·

Methods of expanding tumor infiltrating lymphocytes (TILs), including peripheral blood lymphocytes and marrow infiltrating lymphocytes, from blood and/or bone marrow of patients with hematological malignancies, such as liquid tumors, including lymphomas and leukemias, and uses of such expanded TILs in the treatment of diseases such as cancers and hematological malignancies are disclosed herein.

METHOD FOR INCREASING CONTENT OF NK CELLS USING CULTURE MEDIUM OF IMMUNE CELLS OF HEALTHY PERSON
20220348873 · 2022-11-03 ·

The present invention relates to a culture method for increasing the content of NK cells. According to the present invention, the growth rate of immune cells including NK cells can be increased not only in cancer patients but also in persons who are genetically susceptible to cancer or elderly people, and an NK cell fraction and cell-killing ability can also be enhanced.

TUMOR-INFILTRATING LYMPHOCYTES WITH ENHANCED TUMOR REACTIVITY
20230086675 · 2023-03-23 ·

Disclosed are compositions and methods for targeted treatment of infections and cancers expressing cancers. In particular, tumor infiltrating lymphocytes (TILs) are identified that can be used with adoptive cell transfer to target, penetrate, and kill solid tumor masses. Therefore, also disclosed are methods of providing an immunotherapy in a subject with an infection or cancer that involves adoptive transfer of the disclosed TILs.

COMPOSITIONS AND METHODS FOR DIFFERENTIATING STEM CELLS INTO NK CELLS

The disclosure features methods and compositions for differentiating stem cells into hematopoietic stem and progenitor cells (HSPC) and/or Natural Killer (NK) cells. The methods and compositions described herein are used to differentiate stem or progenitor cells having at least one gene-edit that is maintained in the differentiated cell. Also provided are differentiated cells produced using the methods and compositions described herein for therapeutic applications.

MR1 RESTRICTED T CELL RECEPTORS FOR CANCER IMMUNOTHERAPY
20220339271 · 2022-10-27 · ·

The invention relates to a method of isolating a T cell that expresses a T cell receptor capable of binding specifically to an antigen presented by a cancer cell in association with an MR1 molecule. The method comprises the steps of (a) providing a preparation of T cells, (b) contacting the preparation with cancer cells expressing MR1 protein; (c) isolating a T cell that is specifically reactive to said cancer cells.

The invention further relates to a method of preparing a T cell preparation expressing select MR1 recognizing T cell receptors from transgene expression vectors, the use of such T cell preparations in treatment of cancer, and to collections of MR1 reactive T cell receptor encoding nucleic acids and cells.

MONOCYTE DEPLETION OF T CELLS POPULATIONS FOR T-CELL THERAPY
20230089392 · 2023-03-23 ·

A method for producing an engineered T cell population includes obtaining a cell population containing a monocyte and a T cell, resting the obtained cell population on a surface, adhering the monocyte to the surface, retaining a non-adherent cell population, activating the non-adherent cell population, introducing a nucleic acid into the activated non-adherent cell population to obtain a transformed T cell, and expanding the transformed T cell to obtain the engineered T cell population.
