Adenoviral Vectors
The present invention relates to an adenoviral vector comprising a regulatable Major Late Promoter and an exogenous transgene. The invention also provides cells comprising such adenoviral vectors, and processes using such vectors.
AUTOMATED PRODUCTION OF VIRAL VECTORS
The present disclosure provides an automated method of producing viral vectors, utilizing engineered viral vector-producing cell lines within a fully-enclosed cell engineering system. Exemplary viral vectors that can be produced include lentivirus vectors, adeno-associated virus vectors, baculovirus vectors and retrovirus vectors.
ADENO-ASSOCIATED VIRUS (AAV) PRODUCER CELL LINE AND RELATED METHODS
The present disclosure relates to a mammalian cell line for producing adeno-associated virus (AAV), suitably including nucleic acids encoding helper genes and AAV genes, under the control of derepressible promoters. The disclosure also relates to isolated nucleic acid molecules that encode such genes, as well as methods of using the mammalian cells for producing AAVs.
PROMOTER FOR HETEROLOGOUS EXPRESSION
The present invention is directed to a nucleic acid construct comprising a polynucleotide operably linked to one or more control sequence that directs the expression of the polynucleotide in a host cell, wherein at least one control sequence comprises a promoter sequence of an operon comprising a secA gene or a functional fragment or functional variant thereof and wherein said promoter sequence is heterologous to the polynucleotide. In a further embodiment, the invention is directed to an expression vector and a host cell comprising the nucleic acid construct comprising the promoter sequence described herein and a method of expressing a polynucleotide in a host cell.
PROGRAMMABLE TRANSCRIPTION FACTORS AND METHODS
This disclosure describes a programmable transcription factor system. Generally, the system includes a programmable transcription factor, a polynucleotide that includes a coding region whose expression is either required or toxic for cell viability, and a promoter operably linked to the polynucleotide and having a binding site for the programmable transcription factor. The programmable transcription factor generally includes a domain that specifically binds to a promoter and a domain that either activates transcription (if the polynucleotide encodes an essential product) or represses transcription (if the polynucleotide encodes a toxic product).
METHODS AND SYSTEMS FOR CONDITIONALLY REGULATING GENE EXPRESSION
The present disclosure provides systems, methods, and compositions for conditionally regulating expression of a target gene. Aspects of the present disclosure utilize intracellular signal transduction pathways to regulate the expression of a gene (e.g., transgene, exogenous gene, endogenous gene).
ENGINEERED NK CELLS
Provided herein, inter alia, are compositions including engineered NK cells and methods for preparing the same. The engineered NK cells provided herein include integrated nucleic acid sequences encoding Cas9 proteins (e.g. dCas9). The engineered NK cells are contemplated to be effective for treating and/or preventing cancer, particularly leukemia.
DNA-BASED NEURAL NETWORK
An analog signal processing circuit comprising: a first promoter operably linked to a nucleic acid sequence encoding a first output molecule, wherein said promoter is responsive to a cooperative input signal comprising at least two cooperative inputs, and wherein expression of said at least two cooperative inputs is tunable.
SINGLE-VECTOR TYPE I VECTORS
The invention relates to the production and use of Cas-encoding sequences and vectors comprising these. Aspects of the invention provide products, vectors, delivery vehicles, uses and methods for producing Cas-encoding sequences in bacterial or archaeal cells.
Highly inducible dual-promoter lentiviral TET-ON system
The invention relates to expression systems useful for regulated expression of a gene of interest based on the constitutive expression of the original TetR repressor and the expression of the polynucleotide driven by a constitutive promoter operably linked to an operator sequence for a tetracycline operator sequence. The system can be provided as two different polynucleotides or as an all-in-one vector. The invention also relates to vectors, host cells and viral particles according to the invention as well as to the uses thereof for in vitro and in vivo production of products of interest or for therapy.
