The present disclosure provides RNA-guided CRISPR-Cas effector proteins, nucleic acids encoding same, and compositions comprising same. The present disclosure provides ribonucleoprotein complexes comprising: an RNA-guided CRISPR-Cas effector protein of the present disclosure; and a guide RNA. The present disclosure provides methods of modifying a target nucleic acid, using an RNA-guided CRISPR-Cas effector protein of the present disclosure and a guide RNA. The present disclosure provides methods of modulating transcription of a target nucleic acid.
Single-vector type I vectors
The invention relates to the production and use of Cas-encoding sequences and vectors comprising these. Aspects of the invention provide products, vectors, delivery vehicles, uses and methods for producing Cas-encoding sequences in bacterial or archaeal cells.
COMPOSITIONS AND METHODS FOR TREATING CEP290-ASSOCIATED DISEASE
Compositions and methods for treatment of CEP290 related diseases are disclosed.
DEVELOPMENT OF DUAL-gRNA APPROACH WITH UNDETECTABLE OFF-TARGET EFFECT TO CORRECT C9ORF72 REPEAT EXPANSION AND C9ORF72 PATHOLOGY
A group of gRNAs is provided, and the group of gRNAs comprises a first gRNA molecule; and a second gRNA molecule capable of defining a region in a genome sequence with the first gRNA molecule, wherein the region in the genome comprises a target sequence in need of removal.
Treatment Of Psoriasis With Interferon Induced Helicase C Domain 1 (IFIH1) Inhibitors
The present disclosure provides methods of treating subjects having psoriasis, and methods of identifying subjects having an increased risk of developing psoriasis.
POLYNUCLEOTIDES ENCODING GLUCOSE-6-PHOSPHATASE FOR THE TREATMENT OF GLYCOGEN STORAGE DISEASE
This disclosure relates to mRNA therapy for the treatment of glycogen storage disease type 1a, (GSD-Ia), and related symptoms such as hypoglycemia. mRNAs for use in the invention, when administered in vivo, encode human glucose-6-phosphatase (G6Pase or G6PC), and functional fragments and variants thereof. mRNAs of the invention are preferably encapsulated in lipid nanoparticles (LNPs) to effect efficient delivery to cells and/or tissues in subjects, when administered thereto. mRNA therapies of the invention increase and/or restore deficient levels of G6PC expression and/or activity in subjects. mRNA therapies of the invention further increase the glucose production, and reduce the abnormal accumulation of glycogen and/or glucose-6-phosphate associated with GSD-Ia.
THERMOSTABLE PHYTASE VARIANTS
Provided herein, inter alia, are engineered phytase polypeptides and fragments thereof with improved thermotolerance as well as methods for producing and using the same for enhancing animal performance on one or more metrics.
Compositions and methods for treating pancreatic enzyme deficiencies in mammals
A composition containing enzymes, L-arginine, and other components for treating exocrine pancreatic enzyme insufficiencies and gastrointestinal disorders in mammals. The compositions replace pancreatic enzymes while concurrently treating the side effects of small intestinal bacterial overgrowth and gastrointestinal mucosal degeneration that occurs with pancreatic enzyme loss. The composition can be mixed into animal feeds or sprayed onto extruded feeds prior to consumption.
Compositions and methods of use of CRISPR-Cas systems in nucleotide repeat disorders
The invention provides for delivery, engineering and optimization of systems, methods, and compositions for manipulation of sequences and/or activities of target sequences. Provided are delivery systems and tissues or organ which are targeted as sites for delivery. Also provided are vectors and vector systems some of which encode one or more components of a SIN CRISPR complex, as well as methods for the design and use of such vectors. Also provided are methods of directing SIN CRISPR complex formation in eukaryotic cells to ensure enhanced specificity for target recognition and avoidance of toxicity and to edit or modify a target site in a genomic locus of interest to alter or improve the status of a disease or a condition.
Genome editing system
A system for editing of a target sequence at a locus of a host cell is disclosed. The system has a nucleic acid molecule comprising a nucleic acid segment comprising a targeting RNA sequence and an RNA segment that binds a protein. The system also has a nucleic acid molecule comprising a nucleic acid segment encoding a polypeptide with endonuclease activity fused to a protein that binds the RNA segment. The system also comprises a double stranded DNA molecule comprising DNA comprising at least one nucleotide sequence that is capable of binding to the target sequence at the locus.
